China is rich in resources,and each ethnic group has formed its own medicine system and gradually developed its own medicine system due to its geographical location,customs and habits,etc.Ethnic medicine is also gaining more and more attention.Sabia parviflora is the stems and leaves of Sabia parviflora Wall.ex Roxb.,belonging to the Sabia family,it is a folk medicine of the Buyi and Miao nationalities.Modern research mainly focuses on the chemical components and pharmacological effects.The chemical constituents of the Sabia parviflora mainly include alkaloids,flavonoids,pentacyclic triterpenoids,benzene derivatives,etc.The main pharmacological effects are antioxidant,anti-inflammatory,anti-rheumatoid arthritis,hepatoprotective and other aspects.For the current research on the chemical components of Sabia parviflora,traditional extraction and separation methods are mainly used.However,the traditional extraction and separation is time-consuming and laborious,the chemical components of the traditional Chinese medicine Sabia parviflora are also very complex,the trace components are difficult to separate,and other bioactive components are also unknown,therefore,it is necessary to establish an effective and rapid method to systematically describe the chemical characteristics of Sabia parviflora.So,in order to have a more thorough understanding of Sabia parviflora,the chemical composition of Sabia parviflora was analyzed by ultra performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry(UHPLC-Q-TOF-MS/MS).Select nine chemical components separated by the research team in the early stage as indicators,conduct quantitative analysis on the different origins and different parts of Sabia parviflora,and study the protective effects of the roots,stems and leaves of Sabia parviflora from different origins on liver cell damage,and infer the potential compounds in Sabia parviflora,laying a foundation for the in-depth study of Sabia parviflora.(1)Analysis of Chemical Constituents of Sabia parviflora based on UHPLC-Q-TOF-MS/MSWe used ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry to analyze the chemical composition of S.parviflora.Its chemical components were rapidly identified using the characteristic ion filtration method.A total of 105 components were identified from the alcohol extract of Sabia parviflora,including 44 alkaloids,14 amides,7 benzene derivatives,14 terpenes and 26 other compounds,including 12 potentially novel compounds.The method is fast and accurate,which provides an important basis for the study of active substances of S.parviflora.(2)Determination of nine compounds in S.parviflora by UHPLC-Q-TRAP-MSACQUITY UPLC BEH C18(2.1x100 mm,1.7 μm)Chromatographic colu mn,eluted with 0.1% formic acid aqueous solution(A)-acetonitrile(B)mobi le phase gradient,to establish UHPLC-Q-TRAP-MS for the S.parviflora deter mination of vanillin,Seslignanoccidential A,syringaresinol glucopyranoside,N-t rans feruloyloctamine,N-trans feruloyloctamine,sabianin C,3,4,5-trimethoxyben zoic acid,vanillic acid,and p-hydroxybenzoic acid in S.parviflora.The deter mination method was established and the methodology of the established meth od was investigated.Vanillin in 0.049-9.8 μg/m L(R2=0.9987),Seslignanocciden tial A in 0.00316-1.58 μg/m L(R2=0.9971),syringaresinol glucopyranoside in 0.10834-65.01 μg/m L(R2=0.9985),N-trans-feruloyloctopamine in 0.01064-10.64 μg/m L(R2=0.9964),N-trans-feruloyltyramine in 0.01976-9.88 μg/m L(R2=0.9998),s abianin C in 0.0074-5.92 μg/m L(R2=0.9996),3,4,5-trimethoxybenzoic acid in 0.33608-84.02 μg/m L(R2=0.9951),vanillic acid in 0.99025-99.025 μg/m L(R2=0.9995),p-hydroxybenzoic acid in 0.035411-10.6233 μg/m L(R2=0.9997)concentratio n range have a good linear relationship.Precision of established method,repeat ability RSD ≤ 10%,and recovery rate is in the range of 94%-110%,which laid a certain foundation for the quality control of Sabia parviflora.The result s of thermal map analysis show that the content of N-trans-feruloyltramine in t he roots of Tiane County,Lingyun County,Liucheng County and Luodian Cou nty is high,the content of N-trans-feruloyltyramine in the stems of Tiane Cou nty,Liucheng County,Dewu Town and Luodian County is higher,and the con tent of N-trans-feruloyloctopamine in the stems of Fapa Town is higher,the le aves of Tiane County,Lingyun County,Liucheng County,Dewu Town,Luodia n County,Baishiyan County and Honghe Prefecture have a high content of eu genol glucoside.(3)Study on the liver protection of extracts from different parts of Sabia parvifloraIt was found that the Sabia parviflora has better pharmacological effects in liver protection,previous studies have been done in hepatitis,non-alcoholic hepatitis,this experiment is to explore whether the extracts from different parts of Sabia parviflora have protective effects on the liver.MTT colorimetry is used for the experiment.The research results show that after 24 hours,the root extracts of LO2 cells from Liucheng County,Luodian County,Baishiyan,Nujiang,and Honghe Prefecture showed at 2.5 μg/m L,the cell survival rate increased by about 7% compared to the model group,at 5 μg/m L,the cell survival rate increased by about 20% compared to the model group,reaching 10% μg/m L,the cell survival rate increased by about 27%compared to the model group.After 24 hours,the root extracts of HepG2 cells from Tiane County,Liucheng County,Luodian County,Nujiang River,and Banna showed at 2.5 μg/m L,the cell survival rate increased by about 10% compared to the model group,at 5 μg/m L,the cell survival rate increased by about 20% compared to the model group,at 10 μg/m L,the cell survival rate increased by about 30% compared to the model group.In the future,further research can be conducted on the protective effects of extracts from roots from these regions on the liver.There are different degrees of correlation between the compounds and the survival rate of Sabia parviflora.In particular,there is a significant correlation between Sabianin C and survival rate,it shows that the content of Sabianin C will affect the survival rate of liver cells.Sabianin C may be the liver protective active ingredient of Sabia parviflora.
