Lung cancer is a malignant tumor with high morbidity and high mortality in the world.Non-small cell lung cancer(NSCLC)accounts for about 85%of all lung cancers,and most patients are found at an advanced stage.This study highlights the importance of tumor microenvironment in cancer progression and treatment.In tumor immunity,immune cells play a huge role.This study mainly discusses the adaptive immune cells CD3~+T lymphocytes,CD4~+T lymphocytes,CD8~+T lymphocytes,CD20~+B lymphocytes,CD38~+plasma cells in the tumor microenvironment.The expression of innate immune cells CD56~+natural killer(NK)cells and CD68~+tumor-associated macrophages(TAMs),LCA(CD45,leukocyte common antigen).There was no correlation between the expression of T lymphocyte and B lymphocyte subtypes.The differences of above immune cells in different age,gender,smoking history,pathological type,clinical stage and differentiation degree were analyzed.The relationship between the above immune cells and the prognosis of patients was analyzed.Wax chunks from tumor tissues of 43 patients with NSCLC who underwent lung resection from January 2015 to December 2018 were collected from the Eighth Medical Center of the General Hospital of the People’s Liberation Army.The expressions of LCA,CD3,CD4,CD8,CD20,CD38,CD56 and CD68 in tumor specimens were detected by immunohistochemical method.IBM SPSS Statistics 26.0 software was used to analyze the correlation between T lymphocytes and B lymphocytes in the tumor microenvironment of NSCLC.The relationship between LCA,CD3~+T lymphocytes,CD4~+T lymphocytes,CD8~+T lymphocytes,CD20~+B lymphocytes,CD38~+plasma cells,CD56~+NK cells,CD68~+TAMs and clinicopathological characteristics of patients was analyzed.Kaplan-Meier survival curve was used to make the survival curve.Progression-free survival(PFS)was evaluated by log-rank test.The common immune cell types in NSCLC tumor microenvironment are CD3~+T lymphocytes,CD20~+B lymphocytes,CD68~+tumor-associated macrophages,CD56~+natural killer,less(P<0.0001).In the tumor microenvironment of NSCLC,CD4~+T lymphocytes are positively correlated with CD20~+B lymphocytes,and CD8~+T lymphocytes are positively correlated with CD38~+plasma cells.The infiltrating CD4~+T lymphocytes,CD20~+B lymphocytes,CD38~+plasma cells and CD56~+NK cells in lung squamous cell carcinoma were significantly higher than those in lung adenocarcinoma(P<0.05).The infiltration of CD8~+T lymphocytes in the tumor microenvironment was significantly different in patients over 60 years old and under 60 years old(P=0.032).In NSCLC patients,PFS in the CD68~+tumor-associated macrophage high infiltration group were significantly longer than those in the low infiltration group(m PFS:16.5 months vs 9months,P=0.04),suggesting that high expression of CD68~+tumor-associated macrophages may be associated with good prognosis.In summary,the aim of this study is to investigate the infiltration characteristics of immune cells in the tumor microenvironment of NSCLC,the correlation between different subtypes of T lymphocytes and B lymphocytes,the differences of immune cells in the tumor microenvironment with different clinicopathological characteristics,and their relationship with the prognosis of patients.It provides a theoretical basis for the discovery of predictive factors and biological indicators of NSCLC treatment,and the research of immunotherapy targets,and has certain guiding significance.
