endometrium,eutopic and ectopic endometrium of endometriosis,and to explore the relationship between mismatch repair genes and the occurrence and development of endometriosis,so as to provide theoretical ideas for the pathogenesis of endometriosis.Methods:Patients with endometriosis diagnosed by pathology for the first time in the First Hospital of The First Hospital of Qinhuangdao from January 2021 to June 2022 were selected as the experimental group(32 cases),and ectopic endometrial tissue and eutopic endometrial tissue were collected.And 30 cases of the first operation and pathological diagnosis of uterine fibroids were selected as the control group,and normal endometrial tissues were collected.The expression of hMSH2 and hMSH6 proteins in the eutopic and ectopic endometrium of the experimental group and the normal endometrium of the control group were detected by immunohistochemical method,and the serum CA125 level of the experimental group was detected.The expression of hMSH2 and hMSH6 proteins in different endometrial tissues,menstrual cycle,and r-AFS stage of endomorphosis was analyzed,and correlation between hMSH2 and hMSH6 and serum CA125 levels.To explore the role and clinical significance of hMSH2 and hMSH6 proteins in the occurrence and development of endometriosis disease.SPSS26.0 statistical software was used to perform statistical analysis on the data.Results:1.The positive expression rates of hMSH2 protein in ectopic,eutopic and normal endometrium were 68.75%,59.38% and 40.00%,respectively.There was no significant difference between eutopic and ectopic endometrium(p > 0.05),which was higher than normal entoneum in ectopic and in normal endometrium,and the difference was statistically significant(p < 0.05).2.The positive expression rates of hMSH6 protein in ectopic endometrium,eutopic endometrium and normal endometrium were 53.13%,40.63% and 30.00%,respectively.There was no significant difference between ectopic endometrium,eutopic endometrium and normal endometrium(p > 0.05).3.The positive expression rates of hMSH2 and hMSH6 proteins in ectopic endometrium were 64.28% and 50.00% in secretory phase,72.22%and 50.55% in proliferative phase,respectively.The positive rates in eutopic endometrium were 57.14% and 28.57% in secretory phase,50.00%and 61.11% in proliferative phase,respectively.The positive expression rates of hMSH2 and hMSH6 proteins in normal endometrium were 9.09%and 36.36% in secretory phase,36.84% and 26.32% in proliferative phase,respectively.There was no significant difference in the expression of hMSH2 and hMSH6 between the secretory and proliferative phases in ectopic,eutopic and normal endometrium(p > 0.05).4.Expression in different r-AFS stages of endometriosis: the positive expression rates of hMSH2 protein in eutopic endometrium of stage II,III and IV were 33.33%,63.64% and 61.11%,respectively,and the positive expression rates of hMSH6 protein were respectively 33.33%,36.36 and44.44%;The positive expression rates of hMSH2 and hMSH6 proteins in stage II,III and IV ectopic endometrium were 33.33%,63.64%,77.78%and 33.33%,54.55%,55.56% respectively.There was no significant difference in the expression of hMSH2 and hMSH6 proteins between eutopic and ectopic endometrium(p> 0.05).5.The serum CA125 levels of hMSH2 and hMSH6 positive patients in ectopic endometrium were 99.28±76.03U/ml and 67.62±38.09U/ml,respectively.The CA125 levels of patients with negative expression were48.10±27.28U/ml and 101.06±90.22U/ml,respectively.The serum CA125 levels in patients with positive expression of hMSH2 and hMSH6 in eutopic endometrium were 110.38±76.10U/ml and 109.53±90.45U/ml,respectively.The CA125 levels of patients with negative expression were43.70±25.03U/ml and 65.34±42.71U/ml,respectively.Spearman correlation analysis showed that blood CA125 was positively correlated with the expression levels of hMSH2 protein in both in eutopic and ectopic endometrium(correlation coefficients were rs=0.600,rs=0.398,p<0.05,respectively).and There was no correlation with the expression of hMSH6 protein in the in eutopic and ectopic endometrium(correlation coefficients were rs=0.258,rs=-0.058,p>0.05,respectively).Conclusions:1.Mismatch repair protein hMSH2 may be involved in the pathogenesis of endometriosis,and the abnormal expression of hMSH2 protein in eutopic endometrium may play a key role.2.Mismatch repair protein hMSH6 may not be involved in the pathogenesis of endometriosis.3.The expression of mismatch repair proteins hMSH2 and hMSH6 in eutopic and normal endometrium was not affected by the menstrual cycle.4.The expression of mismatch repair proteins hMSH2 and hMSH6 may not be related to the r-AFS stage of endometriosis.5.The abnormal expression of mismatch repair protein hMSH2 may be one of the reasons for the increase of serum CA125 level in endometriosis patients. | | Keywords/Search Tags: |
