Objective:To analyze the expression of clock gene period 1(PER1)in endometrial cancer and the relationship between circadian rhythm disturbance caused by night/shift work and drug resistance in endometrial cancer.Methods:Clinical specimens of 40 patients with endometrial cancer admitted to the First Hospital of Shanxi Medical University from January 2021 to December 2021 were collected,including 20 patients in the abnormal circadian rhythm group(with history of night/shift work)and 20 patients in the normal circadian rhythm group(without history of night/shift work).The expression levels of the rhythm gene PER1 in endometrial carcinoma were detected by Real-time fluorescence quantitative PCR(RT-qPCR)and Western blot.The expression of P170 resistance protein in endometrial carcinoma was detected by immunohistochemistry.SPSS 26.0 and GraphPad Prism 9.0 software were used for data processing and chart making of RT-qPCR,Western blot and immunohistochemical test results.The expression of PER1 and P170 resistance proteins in endometrial cancer tissues of normal and abnormal circadian rhythm groups was further analyzed,as well as the relationship between circadian rhythm disturbance caused by night/shift work and endometrial cancer drug resistance.Results:1.The expression of PER1 in endometrial cancer tissues was detected by RT-qPCR.Compared with normal circadian rhythm group,PER1 expression in abnormal circadian rhythm group was significantly decreased,and the difference was statistically significant(P<0.05).2.The expression of PER1 in endometrial cancer tissues was detected by Western blot.Compared with the normal circadian rhythm group,the expression of PER1 in the abnormal circadian rhythm group was significantly decreased,with statistical significance(P<0.05).3.Immunohistochemical results showed that the expression of P170 resistance protein in the abnormal circadian rhythm group was higher than that in the normal circadian rhythm group,and the difference was not statistically significant due to the small sample size(P>0.05).Conclusion:Our research group found through previous population epidemiological studies and mechanism studies that the disturbance of circadian rhythm would affect the chemotherapy effect and the survival cycle and quality of prognosis of patients with endometrial cancer.This study further found on the basis of previous studies:Compared with the normal circadian rhythm group,PER1 showed low expression in endometrial cancer patients in the abnormal circadian rhythm group,suggesting that night shift/shift work may reduce the expression of the rhythm gene PER1.The high expression of P170 resistance protein in the abnormal circadian rhythm group suggests that the high expression of P170 resistance protein in night shift/shift induced circadian rhythm disturbance may be one of the mechanisms of drug resistance in endometrial cancer.
