Purpose: To study the effects of propofol on the neuronal apoptosis and expressions of Bcl-2/Bax/caspase-3 in rats of cerebral ischemia reperfusion with qi deficiency and blood stasis syndrome.Method: The subjects of this study are rats with qi deficiency and blood stasis syndrome.The sham operation group,model group,Edaravone group,and three propofol experimental groups(low-dose,medium-dose and high-dose propofol group)were set up respectively,totaling six experimental groups.First,the rat model of qi deficiency and blood stasis syndrome was prepared,and then the rat model of cerebral ischemia reperfusion of qi deficiency and blood stasis syndrome was established.For each experimental group of propofol,propofol was injected from the tail vein of rats according to the standard of low-dose(4mg/kg),medium-dose(10mg/kg)and high-dose(20mg/kg)propofol groups.The rats in the sham operation group and the model group were injected with 4m L/kg normal saline from the tail vein respectively.The rats in the positive drug group were injected with 3mg/kg Edaravone Injection from the tail vein respectively.The rat model of cerebral ischemia reperfusion was established by occluding the middle cerebral artery with thread embolism.Longa scoring method was used to evaluate the neurobehavioral damage of each group of model rats.At the end of the scoring,the rats were killed,the brain tissues of some rats in each group were taken out to make samples.TTC staining was taken to observe the changes of cerebral infarction area,HE staining was used to observe morphological changes of rat cerebral cortical neurons and TUNEL assay was used to detect the degree of apoptosis of nerve cells in the cortex of rats in each group.The brain tissues of the remaining model rats were taken and detected the expression levels of Bcl-2,Bax and Caspase-3 by Western blotting.Result: Compared with the control group,the neurobehavioral scores,cerebral infarction areas,degeneration cell indexes and the number of apoptosis cells of the model group increased significantly(P<0.05).The expression level of Bcl-2 decreased significantly while the expression levels of Bax and Caspase-3 increased significantly(P<0.05).Compared with the model group,the neurobehavioral scores,cerebral infarction areas,degeneration cell indexes and the number of apoptosis cells of low-dose,medium-dose and high-dose propofol groups decreased significantly(P<0.05),the expression of Bcl-2 in brain tissue increased while the expression of Bax/Caspase-3in brain tissue decreased significantly(P<0.05).Conclusion: Propofol could improve the neurobehavioral level of rats and play a role in repairing cerebral nerve dysfunction by reduce effectively the neuronal apoptosis and down-regulate the expression of Bax and Caspase-3 and up-regulate the expression of Bcl-2 of cerebral ischemia reperfusion rats with qi deficiency and blood stasis syndrome.
