Abstract/Summary:
Cholangiocarcinoma(CHOL)is the most common malignant tumor of hepatobiliary system.The present study investigated the clinical significance and biological functions of AL365181.2 in CHOL.The poor survival in CHOL has a probable prognostic molecular marker known as the AL365181.2,and AL365181.2 may be an important regulator of immune cell infiltration.And AL365181.2 may be involved in the regulation of apoptosis-related proteins,impeding the apoptotic process,thereby boost tumor proliferation,and promoting cholangiocarcinoma progress.In this study,we compared the AL365181.2 mRNA expression between CHOL tissues and normal tissues.Our data was collected from The Cancer Genome Atlas(TCGA),the R and Gene Expression Profiling Interactive Analysis(GEPIA)identified the correlation of AL365181.2 with CHOL.Moreover,We carried out CCK-8 experiment,EdU experiment,invasion and migration experiment,scratch experiment,using three cell lines(Hu CCT,QBC399,RBE)to explore the effect of AL365181.2 on the proliferation,cell invasion and migration of CHOL cells.And we use the subcutaneous injection of cholangiocarcinoma cells into the immunocompromised mouse to study cholangiocarcinoma.The present study investigated the clinical significance and biological functions of AL365181.2 in CHOL.AL365181.2 was highly expressed in CHOL,which was associated with tumor regional lymph node metastasis and poor prognosis.Through CCK-8 experiment,EdU experiment,invasion and migration experiment,scratch experiment,we proved the effect of AL365181.2 gene on the proliferation,invasion and migration of cholangiocarcinoma cells.Moreover,AL365181.2 expression level has significant positive correlations with infiltrating levels of Tem cells and negative correlations with infiltrating levels of DC cells.In addition,interleukins,leishmania infection,cell surface interactions at the vascular wall,FC epsilon receptor signaling and immunoregulatory interactions between a lymphoid and a non-lymphoid were differentially enriched in the high AL365181.2 expression phenotype pathway.By subcutaneous injection of cholangiocarcinoma cells into the immunocompromised mouse,we proved that the Growth rate of tumor volume transfected with si-AL365181.2 was significantly reduced compared with the si-NC group.We conclude that the poor survival in CHOL has a probable prognostic molecular marker known as the AL365181.2,and AL365181.2 may be an important regulator of immune cell infiltration.AL365181.2 may be involved in the regulation of apoptosis-related proteins,impeding the apoptotic process,thereby boost tumor proliferation,and promoting cholangiocarcinoma progress.
