Objective:To investigate the effects of different doses of sildenafil citrate on the survival of rat deep epigastric perforator(DEP)flap model.Methods:40 male rats were randomly divided into four groups with 10 rats in each group.Postoperative management:normal saline control group(group A),low-dose sildenafil citrate tablets(group B),medium-dose Sildenafil citrate tablets(group C),high-dose Sildenafil citrate tablets(group D).After the operation,the necrosis of the flap was observed by naked eye,and the flap area was converted into transparent paper 7 days after the operation to calculate the survival rate of the flap.Biopsy samples were collected 7 days after operation,and the pathological changes during the survival of the flap were observed by HE staining.The expressions of vascular endothelial growth factor(VEGF)and CD34 were qualitatively analyzed by immunohistochemistry,and the CD34-labeled microvascular density(MVD)was measured.The activity of superoxide dismutase(SOD),malondialdehyde(MDA)and nitric oxide(NO)were detected by ELISA.Results:1.The survival rate of flap was higher in group B(56.73±1.94)%,group C(69.06±1.55)%and group D(78.73±1.41)%than that in control group A(46.59±1.57)%,and the difference was statistically significant(P<0.05).2.In histological evaluation:the experimental group was associated with reduced inflammation and increased capillary formation compared with the control group.In immunohistochemical evaluation,the positive expression rate of VEGF expression in each experimental group was higher than that in the control group,and the difference was statistically significant.The positive rate of CD34 expression in all experimental groups was higher than that in control group,and the CD34-labeled microvascular density(MVD)values in group B(10.02±0.75),Group C(14.22±0.50)and group D(18.79±1.00)were higher than those in group A(6.85±0.62),with statistical difference(P<0.05).3.The activity of superoxide dismutase in group B(69.97±6.12),Group C(98.00±4.32)and group D(203.26±13.67)was higher than that in group A(47.26±3.03),with statistical difference(P<0.05)compared with the control group.The activity of superoxide dismutase was lower in all groups.The value of malondialdehyde(MDA)in groups B(6.22±0.56),C(3.90±0.34)and D(2.52±0.14)was lower than that in group A(11.11±2.03),and the difference was statistically significant(P<0.05).Nitric oxide(NO)in group B(41.93±2.26),group C(66.62±5.22)and group D(106.57±4.21)were higher than those in group A(29.95±3.30),with statistical difference(P<0.05).Conclusion:Sildenafil citrate could promote the survival of DEP flap in rats,and the dose of sildenafil citrate was dependent on the survival rate of DEP flap in rats.
