Objective:To observe the expression of IRE-1α and GRP78 in intestinal epithelial cells of rats with chronic renal failure(CRF)by Colonic Dialysis with Yishen Formula.Methods:In order to produce a rat model of CRF,45 rats out of a total of 60 healthy male SD rats were chosen at random.The model group was then generated using the adenine gavage method.The normal(N)group consisted of the remaining 15 rats in the experiment.And then,the rats with CRF were randomly divided into three groups: the chronic renal failure(CRF)group,the Niaoduqing colon dialysis(NDQ)group,and the Yishen Formula colon dialysis(YS)group.The YS group was given Yishen Formula colon dialysis,while the NDQ group was given Niaoduqing colon dialysis.The experimental period lasted for four weeks.During the experiment,the changes in the rats’ body weight,serum creatinine(Scr),and blood urea nitrogen(BUN)levels were tracked and recorded.At the end of the fourth week,kidney tissue and colon tissue were collected for HE staining in order to observe histological changes.The expression and distribution of IRE1α and GRP78 in colon tissue of each group was detected and compared by immunohistochemical technique.Real-time fluorescence quantitative PCR was used to determine the m RNA expression of IRE1α and GRP78.Results:(1)The general condition of the rats in the CRF group was worse than that of the normal group,and the weight gain was slowed down(P < 0.05).On the other hand,the general condition of the rats in the NDQ group and the YS group improved in comparison with the CRF group,and their weight increased(P < 0.05).(2)Compared with the N group,the levels of both Scr and BUN in the CRF group were found to be obviously higher(P<0.05).According to the findings of immunohistochemistry,the expression of IRE1α and GRP78 in intestinal epithelial cells belonging to the CRF group was significantly up-regulated(P<0.05).The findings of the PCR experiment showed that both the relative expression of GRP78 and IRE1α m RNA had obviously increased(P < 0.05).(3)Compared with the CRF group,the values of Scr and BUN in the NDQ group and the YS group were reduced(P< 0.05),and immunohistochemistry analysis revealed that the expression levels of IRE1 α and GRP78 in intestinal epithelial cells were downregulated(P< 0.05),and PCR results showed that the relative expression of GRP78 and IRE1α m RNA was significantly reduced(P< 0.05).However,there was not a significant difference in expression between the two groups that were given the treatments(P> 0.05).Conclusions:By regulating the expression of IRE1α and GRP78 through colon dialysis,Yishen Formula is able to preserve the intestinal mucosal barrier and,as a result,improve the renal function of rats with CRF.
