| Objective:To investigate the therapeutic effect and possible mechanism of Buyang Huanwu Decoction combined with BMSCs transplantation on spinal cord traumatic paraplegia.Method:The model of spinal cord injury in rats was prepared by the modified Allen method.Totally 50 Sprague-Dawley rats were stochastically divided into the sham operation group,model group,Buyang Huanwu decoction group(25g/kg),BMSCs transplantation group(1m L BMSCs),Buyang Huanwu decoction+BMSCs group(25g/kg Buyang Huanwu decoction+1m L BMSCs),with 10 rats in each group.In this study,the spinal cord injury model was established by the modified Allen heavy blow method.After 1st,3rd and 5th weeks of continuous administration,the spinal cord function of rats was measured by the modified Tarlov score,the inclined plate test and the cortical somosensory evoked potential wave latency,and the pathological grading of spinal cord injury was observed by HE staining.Immunohistochemical staining was used to detect the number of Br DU-labeled BMSCs in injured spinal cord tissues.Results:1.The model of spinal cord injury in rats was successfully established,and the spinal cord function of rats in the model group was significantly impaired.2.The modified Tarlov score and the skew plate test showed that compared with the sham operation group,the modified Tarlov score and the critical Angle of the inclined plane of the model group were significantly decreased at the 1st,3rdand 5thweek(P<0.05).Compared with the model group,the modified Tarlov scores and the critical Angle of inclined plane in Buyang Huanwu Tang group,BMSCs group and Buyang Huanwu Tang+BMSCs group were significantly increased(P<0.05).Compared with BMSCs group,the modified Tarlov score and the critical Angle of tilt plane of rats in Buyang Huanwu decoction+BMSCs group were significantly increased(P<0.05).3.Cortical somatosensory evoked potential test results showed that the latency of cortical somatosensory evoked potential wave was 17~18ms in the sham operation group.At the 1st,3rdand 5thweek,the latency of somatosensory evoked potential wave in the cortex of model group was significantly increased compared with the sham group(P<0.05).Compared with model group,the somatosensory evoked potential latency of cortex in Buyang Huanwu decoction group,BMSCs group and Buyang Huanwu decoction+BMSCs group was significantly decreased,with statistical significance(P<0.05).Compared with BMSCs group,the cortical somatosensory evoked potential latency of Buyang Huanwu decoction+BMSCs group was lower than that of model group(P<0.05).4.HE staining showed that after the 5thweek of transplantation,the sham operation group showed that the internal structure of spinal cord tissue was clear and complete,and the internal structure of cells was not significantly damaged.In the model group,complete cell structure was rare,gray matter was destroyed and dissolved,gray and white matter were difficult to distinguish,and glial scar was formed.Compared with the model group,the internal structure of the spinal cord tissue in each administration group was clearer,there were smaller necrotic areas in the gray matter,more white matter remained,and the glial scar in the internal structure was significantly reduced.5.Immunohistochemical staining showed that no Brd U-labeled BMSCs positive cells were found in the spinal cord injury area of the sham operation group.Brown and yellow Brd U labeled BMSCs positive cells were observed in the spinal cord tissues of BMSCs transplantation groups.After the 1stweek of BMSCs transplantation,a large number of transplanted cells survived,and a large number of BMSCs still survived in the spinal cord tissue at the 3rdand 5thweek of transplantation.After the 5thweek of transplantation,compared with BMSCs transplantation group,the number of Brd U labeled positive cells in Buyang Huanwu Decoction+BMSCs group was significantly increased(P<0.05).Conclusion:1.Both Buyang Huanwu Decoction group and BMSCs transplantation group have therapeutic effects on rats with spinal cord injury.2.Buyang Huanwu Decoction has promoted the migration of BMSCs.3.Buyang Huanwu Decoction combined with BMSCs transplantation has therapeutic effect on rats with spinal cord injury,and Buyang Huanwu Decoction combined with BMSCs transplantation had better therapeutic effect than BMSCS transplantation alone. |
