| Objective:In this study,data mining system was integrated with Delphi method to scientifically screen dietary formula.On this basis,the extraction process was examined.In addition,rats with dampness resistance and moderate coke syndrome were taken as experimental subjects to systematically explore the effects and mechanisms of various doses of dietary formula on dampness removal of medium coke.At the same time,combined with the quality evaluation,the study of dietary prescription drink formula was carried out to provide a demonstration for the development and research of dispelling dampness therapeutic products.Method:1.Study on prescription rules of medicated diet for improving wet resistance and middle Coke syndrome based on data miningPart of this experiment is based on the treatment of dampness and coke syndrome,and refers to the treatment of dampness disease in Syndrome and Therapy of dampness Disease of Traditional Chinese Medicine,and the medicated diet prescriptions with the corresponding functions are screened and input from the Dictionary of Chinese Medicated Diet.And the collected medicated diet for systematic arrangement,unified treatment.Each item was coded and entered into Access2010 to establish"basic database for Improving wet resistance and Medium Coke Prescription".Statistical software SAS was used to carry out statistical analysis on basic database for improving wet resistance and medium Coke prescription.2.Application of modified Delphi method in diet prescriptionThe rationality of compatibility of data mining formulations was comprehensively evaluated,and 3 data mining formulations were finally selected for addition and subtraction,and a total of 7 formulations were finally formulated by combining common clinical formulations and classical formulations,and experts from several disciplines were consulted by Delphi method for comprehensive decision-making.3.Study on the extraction process of Zhongjiao Dispelling dampness Diet PrescriptionHigh performance liquid chromatography(HPLC)method was established for the detection of each active component in dietary prescription.The optimal extraction process was selected through single factor test and orthogonal test,which laid the foundation for the efficacy evaluation and product development of dietary prescription.4.Efficacy evaluation and related mechanism study of Zhongjiao Dispelling dampness Diet prescriptionEstablish a cold and dampness dampness-medium focal model of"excessive external dampness,improper diet and unsatisfied mood".Forty-eight SD rats were randomly divided into 6 groups:blank group(K),model group(M),positive drug group(Y),low-dose diet group(SL),medium-dose diet group(SM),high-dose diet group(SH,n=8).The doses were 1.1 g/kg(Y)and 4.60 g/kg(SL),respectively.9.20g/kg(SM),18.40 g/kg(SH).The experiment lasted for 6 weeks.The rats in each administration group were given the drug while modeling,and the modeling time was maintained for 6 weeks.5.Effect of dietary prescription on intestinal flora in rats with dampness and moderate coke syndromeThe cecum contents of rats in blank group,model group,positive drug group,and diet formula low-dose,medium-dose and high-dose groups were collected for DNA extraction and detection.The V3-V4 region of 16S rRNA gene was amplified by primers,and then PCR products were purified,quantified and homogenized to construct PE library.The qualified libraries were sequenced by Illumina high throughput sequencing platform.The diversity of intestinal flora of rats in each group was analyzed based on the experimental results,and the composition and abundance of different flora were analyzed at the phylum and genus levels.Meanwhile,the functional prediction analysis of intestinal flora of rats was performed.6.Preliminary development of diet prescription drinksThe additive ratio of dietary prescription beverage was examined,and the sensory evaluation was taken as the index.The single factor test was carried out first,on the basis of which the orthogonal test was used to determine the best formula of dietary prescription beverage,and the electronic tongue system was used to verify the flavor of the best formula.At the same time,the physical and chemical properties and microbial limit of the beverage were tested.Result:1.Among the 1089 Tonic Diet prescriptions counted,the frequency analysis found that 778 kinds of raw materials were involved,including 261 kinds of traditional Chinese medicine.The Chinese medicine with the highest frequency is Zingiber officinale Roscoe,Jujubae Fructus and Dioscoreae Rhizoma,which are all of the same kind of medicine and food.In high-frequency traditional Chinese medicine,the use of tonifying drugs is the highest,and the tonifying drugs rank first.The second order association rule analysis results in 30 relationship combinations,and the third order association rule screening results in 30 association rules.Hierarchical cluster analysis obtained 12 new formulas.2.The positive coefficient of experts in the expert consultation is 100%,and the rationality of seven groups of prescriptions for the treatment of dampness of scorch and the improvement of spleen and stomach is optimized.The final group of prescriptions is Poria cocos 10 g,Radix Astragali 10 g,Radix codonopsis 10 g,Glycyrrhiza 3 g,Amomum villosum 3 g,and Stir fried malt 15 g.3.The results of single factor experiment showed that the effect of decocting solvent on the glycosides of calyx isoflavone and codonopsis in radix astragali was not significant,and even the content of the two components would be reduced.Through orthogonal experiment,the best extraction process was obtained as follows:soak other Chinese medicine except Amomum villosum for 20 minutes,add 8 times of water,decoct for 80 minutes,and extract for 3 times.4.The general physical signs of rats showed that the growth of body mass of model rats gradually slowed down;Spiritual depression,reduced activity,accumulation,laziness,squinting;Dark hair color and other symptoms similar to the syndrome of dampness resistance and middle focus in traditional Chinese medicine.The physical signs of rats have changed after giving various doses of dietotherapy prescription.It is preliminarily believed that the model has been successfully established and the dietotherapy prescription has the effect of removing dampness from the middle focus on the rat model of dampness resistance and middle focus syndrome.Compared with the model group,the content of D-xylose in serum of model rats was significantly increased by high dose of dietotherapy prescription(P<0.01);At the same time,the abdominal circumference index of rats in each dose group of dietotherapy prescription decreased significantly(P<0.01),and the body weight increased significantly(P<0.05).The gastric residual rate of rats in the low and high dose groups of dietotherapy prescription decreased significantly(P<0.01);The content of ADH in serum was significantly decreased in each dose group(P<0.01);The high dose group of dietotherapy prescription can significantly increase the content of GAS and IL-10 in rat serum.The content of TNF-αin serum can be significantly reduced in each dose group(P<0.01).At the same time,the content of MTL showed an increasing trend,but there was no statistical difference.The activity of Na~+-K~+-ATPase in liver of rats in medium and high dose groups was significantly increased(P<0.01).The structure of gastric pits in the gastric mucosa of the model rats disappeared,the glands showed different degrees of atrophy,and part of the jejunum mucosa necrosis and shedding;The gastric pits of rats in each administration group basically recovered,and no obvious pathological changes were found in the jejunum.5.The sequencing results of intestinal flora showed that compared with the blank group,the diversity of flora in the model group decreased,but the species richness index had no significant difference.The composition of species between the blank group and each group is quite different,and there is a certain degree of difference between the high-dose diet and other administration groups.At the phylum level,compared with the blank group,the number of Bacteroides in the model group was relatively increased,and the number of Bacteroides was relatively decreased.The species composition structure of the rat microbial community in each administration group changed.At the genus level,the proportion of Firmicutes in the samples of each group is relatively high.Compared with other groups,the species abundance of the main bacteria in the Actinobacteriota in the blank group has changed,and the trend of the changes of each bacteria in the samples after the intervention of the dietotherapy prescription is similar to that in the blank group.Function prediction analysis shows that the dietotherapy prescription has important functions of metabolism,genetic information processing,and human disease in improving the intestinal flora of rats with damp-resistance syndrome,and the main metabolic pathway functions include lipid metabolism,nucleotide metabolism,energy metabolism,etc.6.The best formula of the additive for the therapeutic prescription is white granulated sugar 8%,vanillin 0.12%,citric acid 0.05%,and the electronic tongue taste intensity radar shows that the best formula of the therapeutic prescription beverage neutralizes the bitter taste of the traditional Chinese medicine,and can achieve a moderate sour and sweet flavor.The quality of the prepared beverage was analyzed.The physical and chemical analysis,coliform group,total bacterial colony and mold detection all met the national health standards.Conclusion:1.The data mining analysis finally obtains some association rules and Tonic Diet prescriptions for improving the syndrome of dampness resistance in the middle energizer,comprehensively considers the efficacy of data mining and the rationality of compatibility,and carries out expert decision-making in combination with the Delphi method.2.Through expert consultation,the best formula was determined.The formula and formula were Poria cocos 10 g,Radix Astragali 10 g,Radix codonopsis 10 g,Glycyrrhiza 3 g,Amomum villosum 3 g,and Stir fried malt 15 g.3.The research part of the extraction process of the dietotherapy prescription gives different index component weight coefficients through multiple index comprehensive scores,making the experimental results more scientific.The experiment has good repeatability and the extraction process is stable and reliable.4.The extract of dietotherapy prescription can improve the body mass of rats with damp-resistance and mid-focus syndrome,and the symptoms of depression,decreased activity,dark hair,easy to fall off,withered and no light.The extract of dietotherapy prescription may play a role in removing dampness by improving gastrointestinal function,water metabolism,Gastrointestinal hormones and immune cytokines,and body energy metabolism.5.The dietotherapy prescription can play the role of removing dampness by changing the structure of intestinal microflora and the abundance ratio of each microflora in rats with the syndrome of dampness resistance and middle coke.The intestinal microflora of the model rats after the intervention of the dietotherapy prescription have recovered to varying degrees,which may further affect the main metabolic pathway functions by regulating metabolism,genetic information,human disease and other functions:lipid metabolism,nucleotide metabolism,energy metabolism,and thus play the role of the middle energizer in removing dampness.6.Through the experiment,the formula of the best flavor dietotherapy drink was analyzed,and multi-dimensional evaluation was carried out to provide reference for the development of the mid-coke damp-removing dietotherapy drink. |
