| Objective:By measuring the expression levels and correlations of FoxP3,TGF-β and IL-10 in endometrial carcinoma tissue,we aim to explore the correlation between three immune molecules and the onset age,FIGO staging,depth of myometrial invasion,histological grading,vascular invasion,and lymph node metastasis of endometrial carcinoma.We aim to explore the possible pathogenesis of endometrial carcinoma and explore the clinical significance of the expression of these three immune molecules in endometrial carcinoma.Methods:Thirty patients with endometrioid carcinoma who were admitted and diagnosed pathologically from May 2021 to February 2023 were selected as the endometrioid carcinoma group.The central tissue of the cancer focus was taken as the cancer tissue and the tissue within 3 cm of the edge of the tumor was taken as the adjacent tissue.Thirty patients who underwent curettage or total hysterectomy due to functional uterine diseases during the same period and were pathologically diagnosed as normal endometrium were selected as the normal control group.Determination of the expression levels of FoxP3,TGF-β and IL-10 in three groups by enzyme linked immunosorbent assay.Use SPSS 26.0 statistical software to conduct statistical analysis on the data,analyze the expression levels and correlations of three immune molecules in cancer tissue,adjacent tissue,and normal endometrial tissue,and analyze the correlation between the three immune molecules and the clinical pathological characteristics of patients with endometrioid carcinoma.Results:1.Compared with the normal endometrium group,the expressions of FoxP3,TGF-β and IL-10 in endometrial carcinoma and adjacent tissues were significantly increased;The expression level of three immune molecules in cancer tissue was significantly higher than that in adjacent tissues;The differences among the three groups were significant(P<0.05).2.Pearson correlation analysis showed that there was a correlation between FoxP3 and TGF-β and IL-10 in endometrial carcinoma tissue,and the expression level of FoxP3 was positively correlated with TGF-β and IL-10 expression levels(r=0.368,P<0.05;r=0.716,P<0.05).3.The expression level of FoxP3 and TGF-β was positively correlated with FIGO stage,histological grade,and lymph node metastasis in patients with endometrioid carcinoma.The later the FIGO stage,the higher the histological grade and with lymph node metastasis,the higher the expression level of FoxP3 and TGF-β(P<0.05),but there was no significant correlation with patient age,depth of myometrial invasion,and vascular invasion(P>0.05).4.The expression level of IL-10 was positively correlated with age,FIGO stage,depth of myometrial invasion,histological grade,and lymph node metastasis in patients with endometrioid carcinoma.Patients with age≥50 years,later FIGO stage,deeper myometrial invasion,higher histological grade,and lymph node metastasis had a higher expression level of IL-10(P<0.05),but there was no significant correlation with vascular invasion in patients(P>0.05).5.The average age of 30 patients with endometrioid carcinoma was(56.07 ± 9.37)years old,while the average age of 30 patients with normal endometrium was(55.87 ± 10.62)years old.There was no significant difference in age between the two groups,and there was no statistical significance(P>0.05).Conclusion:1.FoxP3,TGF-β and IL-10 are all highly expressed in the cancer tissue and adjacent tissues of endometrioid carcinoma,while they are low expressed in normal endometrial tissue,indicating a possible correlation between the three factors and the pathogenesis of endometrial carcinoma.2.In cancer tissue,the expression level of FoxP3 is positively correlated with the expression levels of TGF-β and IL-10(r=0.368,P<0.05,r=0.716,P<0.05).It is speculated that FoxP3 may significantly inhibit the immune response of the body by inducing Treg cells to secrete inhibitory cytokines TGF-β and IL-10,enabling tumor cells to escape surveillance and killing by the immune system,and may be involved in the occurrence and progression of endometrioid carcinoma.3.The relationship between three immune molecules and the clinical and pathological characteristics of endometrioid carcinoma: FoxP3,TGF-β and IL-10 are positively correlated with FIGO staging,histological grading,and lymph node metastasis characteristics of endometrioid carcinoma.The expression level of IL-10 is positively correlated with age of onset and depth of myometrial invasion;However,there was no significant correlation between the expression levels of FoxP3 and TGF-β and the age of onset and the depth of muscle infiltration;There is no significant correlation between the expression levels of the three and the characteristics of vascular invasion.The relationship between the three and the clinical pathological characteristics of endometrioid carcinoma suggests that the three immune molecules may be related to the occurrence and development of endometrioid carcinoma. |
