| Objective To study the effect of allicin on the protein expression profile of human gastric cancer BGC823 cell;To study the signal pathway of allicin induced mitotic catastrophe in gastric cancer BGC823 cells.Methods The in vitro iTRAQ labeling and LC-MS-MS proteomics analysis were used to obtain the protein mass spectrum data of allicin acting on gastric cancer cells.The GO function annotation analysis and KEGG pathway enrichment analysis were conducted for the differential proteins.The IPA online biological platform was used to analyze the protein regulatory network and typical pathway analysis of the differential expression proteins of allicin acting on gastric cancer BGC823 cells.Results 1.Allicin can inhibit the proliferation of gastric cancer BGC823 cells,and make the cell cycle stagnate in G2/M phase and induce mitotic catastrophe.Allicin concentration is 25μmol/L The inhibitory effect on BGC823 cells of gastric cancer was most obvious used for 12 hours.This study selected 25 μmol/L allicin concentration for subsequent proteomic analysis and other experiments.2.Using iTRAQ quantitative proteomics to screen the differential proteins in gastric cancer cell BGC823 after 12 hours of allicin treatment with 25 μmol/L showed allicin in the non-synchronous group could induce the differential expression of 458 proteins in gastric cancer BGC823 cells,among which 85 proteins were up-regulated and 373 proteins were down-regulated;In the synchronous group,allicin could induce the differential expression of127 proteins in gastric cancer BGC823 cells,among which 81 proteins were up-regulated and 46 proteins were down-regulated.3.The results of GO/EKGG enrichment showed that the differentially expressed proteins were mainly enriched in the nucleus by affecting protein binding and metabolism.These differential proteins are mainly involved in the cancer pathway;IPA analysis showed that the pathway of differential proteins to exert biological effects was eIF2α/ATF4 signal pathway.Conclusion 1.Allicin can inhibit the proliferation of gastric cancer BGC823 cells.2.Allicin can induce BGC823 cells to die after mitotic catastrophe.3.Allicin induced mitotic catastrophe in gastric cancer BGC823 cells may be related to eIF2α/ ATF4 signal path. |
