| Objective: To clarify the prescription composition,medicinal material source,medicinal site,dose basis,and processing method of the classic famous prescription Kaixin Powder,clarify the preparation method of the material standard of Kaixin Powder,establish the quality standard of the material standard of Kaixin Powder,and speculate on the material basis and mechanism of action for treating cognitive impairment.Methods: Thin layer chromatography was used to identify four medicinal materials in the material standard of Kaixin San,including Polygala tenuifolia Willd.,Panax ginseng C.A.Mey.,Acorus tatarinowii Schott,and Poria cocos(Schw.)Wolf.High performance liquid chromatography was used to determine the content of Polygala tenuifolia saponin and3,6’-bissinacyl sucrose in the material standard of Kaixin San,and establish its characteristic map.Particle size,water content,and extract content of the material standard of Kaixin San were determined by particle size determination,water content determination,and extract determination methods,respectively.Using databases such as TCMSP,Batman TCM,and ETCM for searching and screening,and referring to relevant literature,a database of active ingredients and their action targets in Kaixin Powder was obtained.Retrieve relevant targets for cognitive impairment in databases such as OMIM and Genecard to obtain a database of disease targets for cognitive impairment.Integrate the active components of Kaixin Powder and disease targets of cognitive impairment to create a Wayne diagram,obtain intersection targets,use Cytascape software to construct an active component disease target network,use String database to construct a protein interaction network of intersection targets,and score to obtain core targets.Perform GO analysis and KEGG analysis on the core targets to speculate on the mechanism of Kaixin Powder in treating cognitive impairment.Molecular docking verification using Autodock software.Results: A TLC identification method was established for four medicinal materials including Polygala tenuifolia Willd.,Panax ginseng C.A.Mey.,Acorus tatarinowii Schott,and Poria cocos(Schw.)Wolf.In the TLC identification,the reference material of Kaixin Powder showed the same color spots or fluorescent spots at the same position as the control medicinal material;A method for the determination of polygala tenuifolia saponins and 3,6’-bissinacyl sucrose was established and the minimum content limit was specified;A feature map method was established,and its similarity was greater than 0.9;The minimum content limits of water and extracts were investigated and specified.Search the component target database and screen 27 active components of Polygala tenuifolia Willd.and 740 corresponding targets;8 active components of Poria cocos(Schw.)Wolf.and 81 corresponding targets;35 active components of Panax ginseng C.A.Mey.and 689 corresponding targets;There are 18 active components of Acorus tatarinowii Schott and 180 corresponding targets.A total of 88 active components and 1534 corresponding targets were obtained.A total of 10659 disease targets related to cognitive impairment were selected by searching the disease target database.A total of 1206 intersection targets were obtained,among which the key active components were:polygala tenuifolia saponin,3,6’-bissinacyl sucrose,and polygalactone III,ginsenoside Rg1,ginsenoside Rb1,ginsenoside Re,etc.Obtain core targets:APP,bcl2,etc.Through GO analysis and KEGG analysis,the main pathways are c AMP signal pathway and calcium signal pathway.Using molecular docking software,the docking binding energy of ginsenoside Rg1 with app was obtained to be<-5k J/mol.Conclusion: The prepared Kaixin Powder material benchmark is uniform and controllable,and the established quality standard can comprehensively control the quality of Kaixin Powder material benchmark.Kaixin Powder may play a role in treating cognitive impairment by regulating related pathways such as c AMP signal pathway and calcium signal pathway. |
