Objective: To study the correlation between Homocysteinne(Hcy)and eclampsia.To investigate Hcy,Folic acid(FA),Vitamin B12(Vitamin B12,Vit B12),Recombinant Placenta Growth Factor(PLGF),and soluble FMS-like tyrosine kinase 1,The relationship between s Flt-1 and the onset of eclampsia and the combined diagnostic efficacy;To establish the methodology of High performance liquid chromatographytandem mass spectrometry(HPLC-MS/MS)method;Enzymatic cycling(ECA)is the Enzymatic Cycling method to evaluate the relevance of HPLC-MS/MS Enzymatic Cycling for determination of serum homocysteine.Materials and Methods: In this study,32 patients with preeclampsia,28 patients with severe eclampsia and 30 healthy pregnant women were selected from the First Affiliated Hospital of Wannan Medical College from March 2021 to June 2022 to extract serum samples from the subjects.Serum homocysteine(Hcy)was determined by HPLC-MS/MS and ECA.The serum samples of the subjects were extracted,and the placental growth factor(PIGF)and tyrosine kinase-1(s Flt-1)in the serum of pregnant women were determined by immunofluorescence method,and folate(FA)and vitamin B12(Vit B12)in the serum of pregnant women were determined by enzymerelated immunosorbent assay(ELISA).Results:In the preeclampsia group,the serum Hcy level in the severe preeclampsia group was higher than that in the healthy control group,and the serum Hcy level in the severe preeclampsia group was higher than that in the preeclampsia group,the difference was statistically significant(P<0.05);In the preeclampsia group,the serum FA level in the severe preeclampsia group was lower than that in the healthy control group,the difference was statistically significant(P<0.01);the serum FA level in the severe preeclampsia group was lower than that in the preeclampsia group,but the difference was not statistically significant(P > 0.05).The serum Vit B12 level of the preeclampsia group and the severe preeclampsia group was lower than that of the healthy control group,with statistical significance(P<0.05);the serum Vit B12 level of the severe preeclampsia group was lower than that of the preeclampsia group,but the difference was not statistically significant(P > 0.05).Serum Hcy level in pregnant women was negatively correlated with FA and Vit B12.In the preeclampsia group,the serum s Flt-1 level in the severe preeclampsia group was higher than that in the healthy control group,and the serum s Flt-1 level in the severe preeclampsia group was higher than that in the preeclampsia group,with statistical significance(P<0.05);the serum PIGF level in the preeclampsia group and the severe preeclampsia group was lower than that in the healthy control group.The serum PIGF level in severe preeclampsia group was lower than that in preeclampsia group,but the difference was not statistically significant(P<0.05).The accuracy evaluation and precision evaluation of the mass spectrometer showed good correlation between HPLC-MS/MS method and ECA method.Conclusion: 1.Mass spectrometry has good precision and accuracy in the determination of plasma Hcy level,and the results are highly correlated with those of ECA method.2.Serum Hcy level was positively correlated with the incidence of PE,but there was no significant correlation between Hcy level and the severity of PE.Serum Vit B12 and FA levels were negatively correlated with Hcy,and Vit B12 and FA were protective factors of PE.3.s Flt-1 level was positively correlated with the occurrence and progression of PE,while Pl GF level was negatively correlated with the occurrence and progression of PE.
