| Objective: To prepare blank nanobubbles,STEAP-1 antibody and salinomycin sodium;to explore the release effect of ultrasonic irradiation on salinamycin-loaded sodium nanobubble drugs;and to construct the transplantation of nude mice with prostate cancer PC3 cells.Methods: Blank nanobubbles and saltomycin-loaded sodium nanobubbles were prepared by thin film hydration and mechanical shock,and targeted nanobubbles containing STEAP-1 antibody were prepared by biotin-avidin,and the size,morphology,distribution and size changes of the three nanobubbles with time were observed under the microscope.;prostate cancer PC3 cells were cultured with different ultrasonic irradiation times(0s,5s,10 s,15s,20 s,25s)and different ultrasonic irradiation intensity(1.0W / cm2、1.5W/cm2、2.0W/cm2、2.5W/cm2).By detecting the viability of nanobubble cells after irradiation,the effect of different irradiation time and intensity on the release of drug-loaded nanobubble drugs was analyzed.Cultivate PC3 cells,take an appropriate amount of subcutaneous injection of cells with logarithmic growth stage,and observe the growth of transplanted tumors.Results: Blank nanobubbles,nanobubbles targeting PC3 cells and sodium saltomycin nanobubbles were successfully prepared,and the three nanobubbles were round,uniform in size and uniformly distributed.Under fluorescence microscopy,the morphology of the targeted nanobubbles was round,the size was consistent,the distribution was uniform,and fluorescence color development was visible around the nanobubbles.Under transmission electron microscopy,uniformly dispersed black granular substances in drug-loaded nanobubbles were considered to be carried salinomycin sodium particles.The concentration of biotinylated nanobubbles was 6.1×108/ ml,the concentration of targeted nanobubbles was 5.2×108 / ml,and the concentration of drug-loaded nanobubbles was 4.6×108 / ml.The average particle size of the blank nanobubble on day 0,day 1,day 2,day 4,day 7,day 10 and day 14 were 190.3±14.6nm,174.3±14.6nm,174.4±14.6nm,177.3±7.6nm,176.0±12.1nm,171.1±12.9nm and 181.3±9.0nm,the size of biotinylated blank nanobubble on day 0 was about 190.3±14.6nm,and the size of nanobubble on day 14 was about 181.3±9.0nm.There was no significant difference between the two(P>0.05)The average particle size sizes of the targeted nanobubbles on day 0,day 1,day 2,day 4,day 7,day 10 and day 14 were 168.7±21.3nm,163.9±28.3nm,185.0±13.3nm,139.7 ±17.0nm,198.2±50.2nm,155.4±6.5nm and 165.8±15.2nm,the size of the targeted nanobubbles on day 0was about 168.7±21.3 nm,and the target nanobubble size on day 14 was about 165.8±15.2 nm.There was no statistically significant difference between the two(P>0.05).The average particle size of the salinamycin-loaded sodium nanobubbles on day 0,day 1,day 2,day 4,day 7,day 10 and day 14 were 134.5±19.7nm,146.6±10.9nm,131.5±5.7nm,133.8±6.9nm,148.3±22.0nm,173.2±16.8nm and 141.1±13.4nm,respectively,the size of drug-loaded nanobubbles on day 0was about 134.5±19.7nm,and the size of drug-loaded nanobubbles on day 14 was about141.3±13.4nm.There was no statistically significant difference between the two(P>0.05).Under the same irradiation intensity,the OD value decreased compared with 0s(P<0.05),and the OD value of 2.5W/cm2 decreased compared with other intensities at the same irradiation time(P<0.05).When the irradiation intensity is 1.5W/cm2,the OD value decreases(P<0.05)compared with 5s in 10s;when the irradiation intensity is 2.0W/cm2,the OD value decreases compared with 10s(P<0.05);when the irradiation time is 10 s,20s and 25 s,the OD value decreases(P<0.05)compared with 1.0W/cm2.At the same irradiation intensity,more sodium salinomycin was released when the irradiation time was 5s,10 S,15s,20 s,and 25 s than in the non-irradiated group.At the same irradiation time,when the irradiation intensity was 2.5W/cm2,more sodium salinomycin was released than that in the 1.0W/cm2,1.5W/cm2 and 2.0W/cm2 groups.Conclusion: Three kinds of nanobubbles were successfully prepared,with uniform bubble size,consistent distribution and good stability.Ultrasound irradiation can help release salinomycin sodium in sodium salinomycin nanobubbles,providing an idea for the treatment of prostate cancer.A nude mouse transplant tumor model of PC3 cells of prostate cancer was successfully constructed,and the transplanted tumor was observed under ultrasound as a low-echoic nodule,the capsule was still intact,the morphology was still regular,the internal echo is uneven,and obvious blood flow signals are visible internally. |
