Objective: It has been determined that endometrial inflammatory factors and progesterone receptor(PR)have recurrent implantation failure,it is expressed in the endometrial tissue of Chronic Endometritis(CE)rat and embryo implantation dysfunction(EID)rat,to explore its effect on RIF and related mechanism.Methods:(1)The secreting endometrial tissues and decidua tissues before 7 weeks of gestation were collected from the Reproductive Center of Yijishan Hospital,Wannan Medical College, and the inflammatory expression of endometrial tissues in the two groups were compared by HE staining. The expressions of inflammatory cytokines CD38/CD138 and progesterone receptor(PR)in endometrial tissues of the two groups were determined by immunohistochemistry.(2)SD rats aged 8-9 weeks were selected to construct the rat model of Chronic Endometritis(CE).After the successful establishment of the animal model, the expression of progesterone receptor(PR)in the endometrial tissues of the two groups of rats was measured by immunohistochemistry.(3)SD rats aged 8-9 weeks were selected to construct the embryo implantation dysfunction(EID)rat model. After the embryo implantation dysfunction(EID)was successfully constructed, the expression of endometrial tissue inflammation in the two groups was compared by HE staining.Western Blot assay was used to determine the expression of inflammatory factors in endometrial tissues of rats in both groups.Results:(1)The positive rate of inflammatory factors was high in RIF population, and endometrial progesterone receptor was abnormal.(2)The fertility of CE model rats decreased and the progesterone receptor was abnormally expressed.(3)The inflammatory factor IL-1β was highly expressed in the endometrial tissue of EID model rats, but the expression of IL-6 and TNF-α did not change significantly.Conclusion:(1)The interaction between inflammatory factors and progesterone receptors in endometrial tissues, and the strong expression of inflammatory factors may lead to antagonism of progesterone receptors, thus reducing fertility.(2)The suppressed expression of progesterone receptors in endometrial tissue may promote the occurrence of inflammatory changes in the uterine cavity, but it has not been confirmed that all inflammatory factors have changed.
