The Role Of LncRNA CASC8 In The Development Of Pancreatic Cancer And The Preliminary Study Of Its Mechanism | | Posted on:2024-04-30 | Degree:Master | Type:Thesis | | Country:China | Candidate:A J Gou | Full Text:PDF | | GTID:2544307088980199 | Subject:Surgery | | Abstract/Summary: | | | Objective:Pancreatic cancer has an insidious onset and atypical early symptoms,and most of patients are already in the middle and late stages when they are clinically diagnosed.Therefore,the search for biomarkers and effective biotherapeutic targets for early diagnosis of pancreatic cancer has been a hot topic of research in recent years.One promising direction is the use of long-stranded non-coding RNA(lnc RNA)as a tumor marker.CASC8 is a lnc RNA discovered in recent years and has been found to play a role in the progression of various tumors.However,little has been reported in the literature about the role of CASC8 in pancreatic cancer.Therefore,in this study,based on the analysis of relevant bioinformatics databases,we found that CASC8 was significantly differentially expressed in most tumors and paired tissues,especially in pancreatic cancer tissues,and was significantly correlated with prognosis.Therefore,studying the effect of CASC8 on pancreatic cancer and its potential as a preventive and therapeutic target may provide a theoretical basis for the treatment of pancreatic cancer.Methods: Bioinformatics databases such as TACG were used to verify the expression levels of CASC8 in human pancreatic cancer tissues compared to paracancerous tissues,the relationship between high CASC8 expression and patient survival and its target genes.At the same time,q RT-PCR was used to verify the difference in expression of CASC8 between the pancreatic cancer cell line and the normal pancreatic epithelial ductal cells.The effects of CASC8 on proliferation,migration and invasion of pancreatic cancer cells and EMT were verified by transfection of sh RNA knockdown CASC8,CCK-8,scratch assay,plate cloning assay,Transwell migration invasion assay and Western blot in pancreatic cancer cell lines As PC-1 and Miacapa-2.The binding of mi RNA to target genes was verified by dual luciferase assay.Results: 1.CASC8 expression is upregulated in pancreatic cancer patient tissues and cell lines and is associated with disease-free survival and poor prognosis.2.CASC8 promotes proliferation,migration and invasion of pancreatic cancer cells in vitro.3.CASC8 affects the EMT of pancreatic cancer cells in vitro,and the knockdown of CASC8 inhibits EMT.4.CASC8 targeting regulates mi R-671-5p expression.5.Reply experiments further validated the targeted regulation of CASC8 to mi R-671-5p.6.mi R-671-5p can inhibit proliferation,migration,and invasion of pancreatic cancer cells in vitro.Conclusion:CASC8 is highly expressed in pancreatic cancer and promotes proliferation,migration,invasion and EMT of pancreatic cancer cells.CASC8 targeted regulation of mi R-671-5p expression,and reply experiments further validated the targeted regulation of CASC8 with mi R-671-5p.Validation of mi R-671-5p on proliferation,migration and invasion of pancreatic cancer cells.Thus,the regulatory mechanism of long-stranded noncoding RNA CASC8 in pancreatic cancer was established. | | Keywords/Search Tags: | CASC8, pancreatic cancer, proliferation, migration, invasion | | Related items |
