Purpose:Fucoidan is a natural extraction with many biological activities.Different origin of fucoidan has different molecular wight,and will cause different change in immune system.It was recently reported fucoidan could inhibit inflammation by affecting cell signal pathways,including mitogen-activated protein kinase(MAPKs),Signal transducer and activator of transcription(STAT)pathway,and Phosphoinositide-3-kinase/AKT serine-threonine kinase/mechanistic target of the rapamycin kinase(PI3K/AKT/m TOR)pathway.Primary immune thrombocytopenia is an acquired auto-immune disease.The pathways mentioned above were reported pathogenic in ITP.Therefore,fucoidan might alleviate the disease activity of ITP according to the literature.To prove that a specific kind of fucoidan can inhibit the activity of MAPKs,STAT,PI3K/AKT/m TOR pathway.To find the immune modulatory effect of the fucoidan on ITP patients immune cells.To discuss a possible mechanism of the fucoidan’s immune modulatory role.Methods:First,the molecular wight of the fucoidan obtained from???was determined by mass spectrometry.Then the specific fucoidan was used to treating peripheral blood mononuclear cells(PBMCs)from ITP patients and a cell line 549 in vitro.Flowcytometry analysis,real-time polymerase chain reaction,and Western Blot were used to identify the effect of this fucoidan.py MOL software was used to demonstrate the possible mechanism of fucoidan on immune system.Results:Fucoidan changed the percentage of ITP patients’CD4~+T cells subtypes and macrophage subtypes,showed by flowcytometry analysis.In cell line A549 tests,fucoidan upregulated the expression of interleukin-6(IL-6)and IL-17,downregulated the expression of IL-10.Fucoidan could also decrease the phosphorylation of MAPKs,STAT,PI3K/AKT/m TOR kinases.The combination of fucoidan and its candidate ligands was demoed by py MOL software,which provided a new tool to learn how did the fucoidan work inside cells.Conclusion:The fucoidan used in this study had an immunomodulatory effect on ITP patients’PBMCs.The fucoidan might decrease the phosphorylation of MAPKs,STATs,and PI3K/AKT/mTOR pathways kinases by combining with FGF1.Fucoidan was a potential drug used in treating ITP.
