Objective:To study the cytotoxicity and proliferation properties of electrospinning.Methods:Osteoblasts and vascular endothelial cells of rats were cultured in vitro,and the electrospun PVDF bionic periosteum was cultured with osteoblasts and vascular endothelial cells respectively.The experimental groups were as follows:PVDF group was bionic periosteum without adding any metal ions.PVDF-Zn group is the bionic periosteum with Zn2+added in the process of electrospinning.PVDF-Mg group is the bionic periosteum with Mg2+added in the electrospinning process.PVDF-Zn-Mg group was the bionic periosteal with Zn2+and Mg2+added during electrospinning.The cell morphology was observed by gross observation,osteoblast alkaline phosphatase staining,vascular endothelial cells CD31immunofluorescence staining,scanning electron microscopy to observe the adhesion of bionic periosteal surface cells,and the absorbance value of cells in each group was detected by CCK-8 method.The relative cell proliferation rate was calculated according to the formula to evaluate the cytotoxicity of electrospun PVDF bionic periosteum.Results:Through gross observation of cell morphology,alkaline phosphatase staining of osteoblasts and immunofluorescence staining of vascular endothelial cells,the results showed that the four groups of piezoelectric biomimetic periostium were conducive to cell adhesion,extension and proliferation during co-culture.Scanning electron microscope observed the cell adhesion experiment,the bionic periostium cells in each group were fully spread,the growth shape was good,and the cells were polygonal.It is firmly covered on the surface of the fiber,and the cellular pseudopod extends to the periphery of the fiber membrane and the void,showing good cellular compatibility.The results of the cytotoxicity experiment of CCK-8 cells showed that the relative proliferation rate of CCK-8 cells was≥75%when co-cultured with osteoblasts,and the cytotoxicity grade was≤1.According to the cytotoxicity grading standard,when the cytotoxicity grade was≤1,there was no cytotoxicity.When co-cultured with vascular endothelial cells,the relative proliferation rate was≥125%,and the cytotoxicity was grade 0.Conclusion:Electrospinning PVDF group,PVDF-Zn group,PVDF-Mg group and PVDF-Zn-Mg group have no cytotoxicity and good cytocompatibility.
