Objective:To study the chemical constituents of the dried roots of Rhodiola crenulata(Hook.f.et Thoms.)H.Ohba,a plant of the Sedumaceae,and to investigate the anti hypoxic damage effects of the chemical constituents from Rhodiola crenulata.Methods:The chemical components in Rhodiola were systematically isolated and purified using silica gel,MCI,ODS,Sephadex LH-20,and HPLC chromatography methods.The structure of the isolated chemical components was identified using advanced spectral and spectroscopic methods such as UV,IR,NMR,and MS.The protective effect of the isolated compounds on Co Cl2induced PC 12 cells was evaluated using the CCK-8 method.Results:60 compounds were isolated and identified from the 70%ethanol extract of Rhodiola.It includes 19 phenylpropanoids,14 phenols,7 phenylethanes,7 fatty chains,4flavonoids,and 9 others.Among them,compounds 1 and 2 were new compounds,compounds 9,12,19,20,21,23,24,32,43,47,and 58 are isolated from the genus Sedum for the first time,and compounds 6,8,33,46,48,50,54,55,56,57,59,and 60 were isolated from the genus Sedum for the first time.At 20μmol/L,phenylpropanoid,phenols,phenylethanes,and flavonoids all showed significant anti hypoxic damage effects.Conclusions:This experiment systematically studied the chemical components of Rhodiola and conducted in vitro anti hypoxic injury activity experiments on the isolated compounds.This study laid the experimental foundation for the pharmacological components of Rhodiola and provided theoretical basis for its development and utilization...
