| Objective:By observing the anti-fatigue effect of Tianxing Jianshenwei Granules on D-galactose induced aging model rats,the anti-fatigue mechanism of Tianxing Jianshenwei granules was discussed.Methods:A total of 60 6-week-old male SD rats were selected and fed adaptively for one week.According to random number table method,the rats were randomly divided into 6 groups: blank group,model group,vitamin E positive control group,Tianxing Jianshenwei granule high-dose,medium-dose and low-dose groups,with 10 rats in each group.The blank group was given daily intraperitoneal injection of normal saline for six weeks.The aging rat model was induced by intraperitoneal injection of D-galactose at the injection dose of 0.5g/kg in the other 5 groups,and the total modeling time was 42 days.Starting from the 15 th day of modeling,vitamin E group was given 27mg/kg vitamin E solution;Tianxing Jianshenwei granule high,medium and low dose groups were given 10.8g/kg,5.4g/kg and 2.7g/kg Tianxing Jianshenwei granule solution;blank group and model group were given equal volume normal saline.They were weighed once a week,and the gavage dose was adjusted according to the weight after weighing,and gavage was administered for 28 days.The general state of the rats was observed during the modeling process.After the modeling,the difference of exercise endurance of the rats was compared through exhaustive swimming experiment.The contents of serum urea nitrogen,blood lactic acid,lactate dehydrogenase,liver glycogen and muscle glycogen in rats were detected after exercise.HE stained sections of rat skeletal muscle and liver were prepared to observe the changes of morphological structure.Results:1.Compared with the blank group,the overall state performance of rats in the model group was poor,with dry,rough and loose skin,yellow hair,lack of luster and sparse hair accompanied by severe shedding.Often huddle,activity reduced;Compared with model group,the rats in vitamin E group and Tianxing Jianshenwei granule administration groups recovered well,acted quickly,had better mental state,and had the same activity frequency,aggressive and active.2.Compared with the blank group,the load-bearing swimming time of rats in the model group was significantly reduced;Compared with model group,the swimming time of rats in vitamin E group and Tianxing Jianshenwei granule groups with different doses was increased in different degrees.The improvement of weight-bearing swimming time was most obvious in Tianxing Jianshenwei granule high-dose,medium-dose and vitamin E groups.3.Compared with blank group,the content of serum urea nitrogen and blood lactic acid in model group increased;Compared with model group,the contents of serum urea nitrogen and blood lactic acid decreased in different doses of Tianxing Jianshenwei granules.The content of serum urea nitrogen and blood lactic acid in Tianxing Jianshenwei granule high-dose group decreased most obviously.Compared with blank group,lactate dehydrogenase activity decreased in model group.Compared with model group,the lactate dehydrogenase activity of Tianxing Jianshenwei granule groups was enhanced.Among them,the activity of lactate dehydrogenase in Tianxing Jianshenwei granule high-dose and medium-dose groups was the most obvious,and the content of lactate dehydrogenase in vitamin E group was increased,with no statistical significance.4.Compared with blank group,the content of liver glycogen and muscle glycogen in model group decreased.Compared with model group,the contents of liver glycogen and muscle glycogen in vitamin E group and Tianxing Jianshenwei Granule high-dose and medium-dose groups increased significantly after exercise,and the effect was most obvious in Tianxing Jianshenwei granule high-dose group,while there was no statistical significance in Tianxing Jianshenwei granule low-dose difference.5.Compared with blank group,liver tissue and skeletal muscle tissue cells in model group showed obvious inflammatory damage and skeletal muscle fiber deformation.The structure of liver tissue and skeletal muscle histiocyte of rats in each administration group of Tianxing Jianshen Granules were improved to varying degrees.Conclusion:1.Tianxingjian ginseng flavor granules can improve the survival-like state of D-galactose-induced aging rats to a certain extent,prolong the weight-bearing swimming time of rats,and improve the morphological structure of skeletal muscle and liver tissue.2.Tianxingjian ginseng flavor granules can enhance the activity of lactate dehydrogenase in D-galactose-induced aging rats to a certain extent,reduce the accumulation of lactic acid in muscles after exercise,inhibit the production of urea nitrogen,improve the level of liver glycogen and muscle glycogen,and delay the occurrence of fatigue.Its mechanism of action may be related to regulating the metabolism of energy substances,accelerating energy substance reserves,and reducing inflammatory damage.In short,Tianxingjian ginseng flavor granules have good anti-fatigue effect. |
