The Effect Of Long Non-coding RNA DLGAP1-AS1 On The Migration,Invasion And Proliferation Of Osteosarcoma Cells | | Posted on:2024-03-20 | Degree:Master | Type:Thesis | | Country:China | Candidate:Q Z Zhao | Full Text:PDF | | GTID:2544307082969599 | Subject:Surgery (bone) | | Abstract/Summary: | PDF Full Text Request | | Objective Osteosarcoma(OS)accounts for about 56 % of orthopedic-related tumor diseases.It is one of the most common malignant bone tumors in orthopedics.It often occurs in children and adolescents,especially those aged about 16 years.At present,the standard treatment and nursing methods for OS include extensive surgical resection and adjuvant chemotherapy.However,according to relevant literature reports,OS has a high local invasion ability and a tendency to early metastasis,and clinical studies have shown that OS will be transferred to the human lung first,causing substantial lesions;approximately 30-50 % of people suffering from the disease have recurrent disease during follow-up;approximately 68 % of patients with localized OS survive less than 5 years or longer.Several anticancer drugs have been developed clinically,but the treatment and prognosis of metastatic and recurrent OS have not been well controlled in the past few decades.In view of these obstacles in traditional treatment regimens,it is urgent to develop new treatment strategies to improve the overall survival rate of OS patients.Long non-coding RNA(lnc RNA)is an important regulator in the gene expression network.They control the nuclear structure and transcription in the nucleus and regulate the stability,translation and modification of m RNA in the cytoplasm.At present,lnc RNA is rapidly becoming a new class of transcripts related to various cellular and biological processes.They are abundantly expressed and widely associated with a variety of cancers.Abnormal expression and mutation are closely related to tumorigenesis,metastasis and tumor staging.Therefore,we tried to construct a gene prognostic signature of osteosarcoma-related lnc RNA to predict the treatment and prognosis of osteosarcoma patients and provide a new direction target for the treatment of osteosarcoma.Methods(1)We first used q RT-PCR(reverse transcription-polymerase chain reaction)to detect the expression of lnc RNA DLGAP1-AS1 in normal human bone osteoblasts(h FOB1.19)and osteosarcoma-related cell lines(143B,MG-63,R-1059-D,HOS).(2)The appropriate osteosarcoma cells 143 B and MG-63 were screened according to the PCR in(1).Then 143 B and MG-63 were transfected with si RNA DLGAP1-AS1,and the transfection efficiency of si RNA was verified by q RT-PCR.The effect of DLGAP1-AS1 on the proliferation of osteosarcoma cells was detected by CCK-8 assay.(4)The migration and invasion ability of osteosarcoma cells were detected by Transwell assay and biofilm assay.(5)The effect of DLGAP1-AS1 on the expression of epithelialmesenchymal transition(EMT)-related proteins in osteosarcoma cells was detected by Western-Blot(WB).Result(1)Compared with normal osteoblasts,the expression of lnc RNA DLGAP1-AS1 in some osteosarcoma cells was increased.(2)The lnc RNA DLGAP1-AS1 plasmid was successfully transfected and the silencing DLGAP1-AS1 osteosarcoma cell model was constructed.(3)Silencing of lnc RNA DLGAP1-AS1 can inhibit the migration,invasion and proliferation of osteosarcoma cells.(4)The silencing of DLGAP1-AS1 can reduce the expression level of epithelial-mesenchymal transition(EMT)related protein NCadherin and ZEB1.Conclusion In this study,in vitro cell experiments verified that silencing DLGAP1-AS1 expression in osteosarcoma cells can inhibit the proliferation,migration and invasion of osteosarcoma cells.Therefore,DLGAP1-AS1 may become a new target for the treatment of osteosarcoma. | | Keywords/Search Tags: | Osteosarcoma(OS), Long non-coding rna(lncRNA), DLGAP1-AS1, Proliferation, Migration, Invasion | PDF Full Text Request | Related items |
