Study On The Correlation Between The Expression Of Melatonin Receptor MT2 And Oxidative Stress Response In Placenta Of GDM Pregnant Women

Objective:The oxidative stress markers Heme Oxygenase1(HMOX1&HO-1)and Nuclearfactor E2 related factors 2(nuclear Factore2-related Factor 2)in placenta treated with GDM through clinical data analysis and experiments were investigated.Nrf2)and melatonin receptor 1B(MTNR1B,MT2),to further explore the correlation between the level of melatonin receptor and the content of oxidative stress markers in the placenta of GDM pregnant women.Method:A total of 84 pregnant women who met the inclusion criteria were selected,including 38 in the GDM group and 46 in the control group.Relevant clinical data and fresh placental tissue during delivery were collected.The expressions of oxidative stress markers heme oxygenase-1(HO-1),nuclear factor E2-related factor 2(Nrf-2)and melatonin receptor 1B(MT2)in placental tissues were determined by middle Westernblot and immunohistochemical staining.Results:There was no difference between GDM pregnant women and normal pregnant women in terms of delivery mode,giant birth rate and incidence of neonatal hypoglycemia(P>0.05).The incidence of neonatal hyperbilirubinemia in GDM group was higher than that in control group(P<0.05).Westernblot and immunohistochemistry showed that the expressions of HO-1,Nrf-2 and MTNRIB in placenta of pregnant women in GDM group were higher than those in control group(P<0.05).The content of MTNR1B in placenta was positively correlated with the content of Nrf-2 in placenta,but the correlation with the oxidative stress marker HO-1 was not clear.Conclusions:1.The level of oxidative stress in GDM pregnant women was higher than that in normal blood glucose pregnant women;2.2.The content of MTNR1B in placenta of GDM pregnant women was higher than that of normal pregnant women;3.There is a certain correlation between placental MTNR1B level of GDM pregnant women and the content of NR-2,a marker of oxidative stress.No relationship has been found between placental MTNR1B level and oxidative stress response of pregnant women with normal blood glucose.4.Pregnancy weight gain of normal pregnant women is positively correlated with newborn weight,while pregnancy weight gain of GDM pregnant women is not significantly correlated with newborn weight.Pre-pregnancy BMI of normal weight mothers has little influence on newborn weight.
