Objective: To explore the expression and clinical significance of pyroptosisrelated genes in patients with bladder cancer.Method: Through online analysis websites and the transcriptome data of bladder cancer and the corresponding clinical data obtained from the TCGA database,we analyzed the differential expression of pyroptosis-related genes between bladder cancer and normal tissues and prognostic value of pyroptosis-related genes in bladder cancer,explored the gene changes and co-expression of differential genes,analyzed the enrichment of differential genes to help understand their biological processes,which provided a basis for further exploration of molecular mechanisms,explored the correlation between differential genes and tumor-related pathways and drug sensitivity,and analyzed the correlation between the expression level of differential genes and the immunocyte infiltration abundance,the expression level of immune molecules and the gene expression of immune checkpoints,so as to provide potential therapeutic targets and prognostic factors for bladder cancer patients in clinical practice.Results: A total of 19 pyroptosis-related genes were identified with differential expression in bladder cancer and normal tissues in this study.It was found that the prognosis of the group with genetic changes in differential pyroptosis-related genes was better than that of the group without genetic changes.In addition,three genes(AIM2,CASP9,GSDMB)related to the prognosis of bladder cancer patients were selected through COX regression and LASSO regression to construct a risk score prognostic model,which was confirmed to be an independent prognostic factor related to the overall survival of bladder cancer patients.At the same time,it was found that these three pyroptosis-related genes were related to some clinical characteristics of bladder cancer patients.Differential pyroptosis-related genes could activate or inhibit tumor-related pathways,and the expression of some genes was found to be related to the drugs’ sensitivity of bladder cancer cells.What’s more,the most of the differential pyroptosis-related genes’ expression was related to the abundance of infiltration of neutrophils and dendritic cells.And there was a strong correlation between the expression of AIM2,IL6,NLRP3 and T cells infiltration abundance and the expression of immune molecules and immune checkpoints genes.Above all,those findings might provide new predictors for the drug efficacy of bladder cancer and provide potential new directions for the selection of drug therapy targets of bladder cancer.Conclusion: The pyroptosis-related genes have shown certain therapeutic and prognostic value in bladder cancer patients,providing more data for the therapeutic targets and efficacy prediction of bladder cancer. |
