| Objective: To explore the role of heat shock protein 70(Heat Shock Protein,HSP 70)in myocardial ischemia and reperfusion injury(Myocardial ischemia reperfusion injury,I/R)in rats with Sanguis Draconis flavones(SDF),and to explore the potential mechanism of total flavonoids on myocardial ischemia and reperfusion injury.Methods: During the experiment,40 male SD rats(around 250g)were used to tie the left anterior coronary artery on the test table: normal group(group Control),sham group(Sham group),myocardial ischemia / reperfusion group(group I/R),and I/R + total flavonoids pretreatment group(I/R + SDF group).A myocardial I/R model was established by reperfusion 120 min after 30 min of left descending anterior coronary artery flow occlusion in body ligation in SD rats.Sham group: opening but without ligation at 1 / 3-2 / 3 below the left anterior coronary artery.Group Control,Group Sham and Group I/R: 14 days before I/R modeling,1 time a day with a volume of 10 m L kg-1.I/R + SDF group: 18 mg/m L of SDF suspension was administered 14 days before I/R modeling,and the time and volume were the same as the other groups.After 120 min of reperfusion,rat abdominal main artery blood and myocardial tissue below the ligation line were taken for relevant index detection.The ECG of each groups were recorded after ischemia and reperfusion,Measure the ST segment changes;The measurement of CK-MB and LDH in rat serum;myocardial histopathorphology of rats was observed by HE staining;Hematoxylin-Eosin(Hematoxylin-Eosin,HE)staining for the observation of myocardial histopathomorphology;Triphenyl tetrazole chloride(Triphenyl Tetrazolium Chloride,TTC)staining to observe the area of myocardial infarction;Quantitative real-time polymerase chain reaction(Quantitative Real-Time PCR,RT-PCR)to detect the HSP70 m RNA expression.Results:(1)Using a method of reciprocal comparison between pairwise groups,Comparing the Control group to the Sham group,The activity and composition in CK-MB and LDH,the degree of myocardial tissue lesion and the degree of necrosis of the morphology and the area of myocardial infarction(P> 0.05);Using the method of mutual comparison between the respective groups,After comparing the Sham and Control groups with the I/R groups,Blood activity in CK-MB,LDH was very active(P <0.05),The pathological changes of the degree of myocardial tissue lesions can be seen in the field of view(HE staining results),Typical necrosis of myocardial ischemic body surface area and expansion of myocardial infarction area(P <0.05)(TTC staining results);SDF pretreatment significantly decreased the serum pathological changes of the degree of concentration of CK-MB,LDH in I/R rats(P <0.05),Significant antly reduced myocardial infarction area(P <0.05),Histopathological damage in declining myocardial I/R rats.(2)Compared The analysis of protein expression data of HSP70 m RNA was not significant(P > 0.05);HSP70 m RNA and method of mutual comparison protein expression in I/R group(P <0.05);HSP70 m RNA and protein expression in I/R + SDF group(P <0.05)in cardiomyocytes(P <0.05).Conclusion:(1)Pretreatment with Total flavonoids can protein expression data of reduce myocardial ischemia and reperfusion injury in rats.(2)Total flavonoids may act to reduce I/R injury in rats by affecting the expression of myocardial HSP70 m RNA downstream or upstream of unknown signaling pathways. |
