Abstract/Summary:
Objective: Kinesin Family Member 20B(KIF20B),thememberofthekinesinssuperfamily(KIFs).KIF20 B was involved in multiple cellular processes such as cytokinesis and axonal transport.Studies have found that the abnormal expression of KIF20 B promotes the occurrence of tumors such as bladder cancer,hepatocellular carcinoma and colorectal cancer.The expression of KIF20 B in gastric cancer(GC)is unknown and its role in the progression of GC is unclear.Microsatellite Instability(MSI)is the absence of Mismatch Repair Protein(MMRP),Few studies on the clinicopathological features and prognosis of P53 and MSI in GC have been reported.To investigate the expression of KIF20 B in low-grade intraepithelial neoplasia(LGIN),high-grade intraepithelial neoplasia(HGIN),GC tissues and corresponding adjacent tissues;to investigate the expression of P53 and MSI;and to explore the clinicopathological features and prognosis of KIF20B、MSI and P53 in GC;and analyze the relationship between KIF20 B.Methods:(1)From April 2019 to June 2022,112 patients data with GC were collected in The Affiliated Hospital of Youjiang Medical College for Nationalities;(2)From January 2020 to December 2021,28 patients with LGIN,32 patients with HGIN underwent endoscopic resection in hospital;(3)Immunohistochemical staining was used to detect the KIF20 B expression in GC,LGIN,HGIN and adjacent tissues;P53 expression in GC and adjacent tissues was detected;MMRP expression in GC tissues was collected.(4)KIF20B expression in GC,LGIN,HGIN and adjacent tissues were statistically analyzed;P53 expression in GC and adjacent tissues were statistically analyzed;MMRP expression in GC tissues were statistically analyzed;(5)The correlation between KIF20 B,p53 and MSI was analyzed.(6)Results from Kaplan-Meier survival analysis,single variable and multivariable Coxregression model analyses showed that the relationship between KIF20 B,P53 and MSI in prognosis.(7)Analysis of KIF20 B expression in gastric cancer tissues and analysis prognosis using online databases.Results:(1)KIF20B expression in GC was higher than that in LGIN,HGIN and adjacent tissue,and KIF20 B expression in HGIN was higher than that in adjacent tissues,P53 expression in GC was higher than that in adjacent tissues(P < 0.05).MLH1 is associates with PMS2,MSH2 is associates with MSH6.(2)KIF20B expression was correlated with depth of invasion,vascular tumor thrombus,lymph node metastasis and nerve invasion;P53 expression was correlated with intestinal metaplasia,sex,depth of invasion and lymph node metastasis,MSI was correlated with depth of invasion,lymph node metastasis and vascular tumor thrombus(P < 0.05).(3)The poor prognosis associated with the high expression of KIF20 B and P53;The favorable prognosis associated with MSI.(4)Results from Kaplan-Meier survival analysis,single variable and multivariable Coxregression model analyses showed that tumor diameter,lymph node metastasis and MSI were independent prognostic factors.(5)Correlation analysis showed that KIF20 B in GC tissues was positively correlated with P53;MSI and P53 expression were negatively correlated,MSI and KIF20 B expression were negatively correlated.(6)In the online database,KIF20 B expression was highly expressed in STAD tissues(P < 0.05),the expression of KIF20 B had no influencedon prognosis of GC patients.Conclusion:(1)KIF20B expression in GC and HGIN was higher than that in adjacent tissue,and KIF20 B expression in GC was higher than that in LGIN,HGIN,The high expression of KIF20 B is involved in the occurrence,invasion and metastasis of GC and may be a prognostic marker of GC.(2)The expression of KIF20 B,P53 and MSI in GC may be regulated by each other.
