Objective: To investigate the relationship between SNP at rs9914220,rs4969168 and rs4969170 of SOCS3 gene and the development of asthma in Guangxi Heiyi Zhuang Children.To analyze the distribution differences of genotype and allele frequency distributions of these three loci rs9914220,rs4969168 and rs4969170 between Heiyi Zhuang nationality in Guangxiand other ethnic and regional areas.Methods: In the case-control study,100 Guangxi Heiyi Zhuang children who were diagnosed with asthma at the Affiliated Hospital of Youjiang Medical University for Nationalities from Oct 2020 to Oct 2022 were selected as the asthma group,and 100 healthy Guangxi Heiyi Zhuang children who checked up at the same time period were selected as the healthy group.Genotypes of rs99140220,rs4969168 and rs4969170 of SOCS3 gene were detected by ligase detection reaction.The Hardy-Weinberg Equilibrum(HWE)law was used to test the population representativeness of the samples.The data was unpacked by SPSS 27.0statistical software.The genotype and allele frequencies of two groups were calculated by manual count method.Chi-square test or Fisher exact probability method was used to compare the genotype distribution and allele distribution between asthma group and healthy group.The logistic regression was used to analyze the risk of genotypes and alleles.The risk of asthma was expressed as a ratio(OR)and a 95% confidence interval(95%CI).The statistical analysis showed that P< 0.05 was considered statistically significant.Results:(1)Comparison of general data: In the asthma group,the average age of children was(4.14±2.24)years old,and the average weight was(15.68±4.76)kg,including 70males(70.0%)and 30 females(30.0%).In the healthy group,the average age of children was(4.06±1.76)years old,and the average weight was(15.78±3.11)kg,including 60 males(60.0%)and 40 females(40.0%).There was no significant discrepancy between two groups in gender,weight and age(P>0.05).(2)SOCS3 gene rs9914220,rs4969168 and rs4969170 in both the asthma group and the healthy group were in line with HWE law.(3)Three genotypes CT,TT and CC were detected at rs9914220 of SOCS3 gene in the asthma group and healthy group of Heiyi Zhuang nationality.Two alleles were C and T.The genotype frequency in the asthma group was CT 39.0%,TT 15.0% and CC 46.0%.The allele frequencies of C and T were 65.5% and 34.5%.The genotype frequency in the healthy group was CT 40.0%,TT15.0% and CC 45.0%.The allele frequencies of C and T were 65.5% and 34.5%.There was no statistic discrepancy in gene frequency and allelic frequency at this locus between the two groups(P>0.05).Comparison of the data in the rs9914220 locus with the ensemble database yielded that the genotype frequency and the allele frequency of this locus in the Heiyi Zhuang population was significantly different from that in Americans,Europeans and South Asians(P<0.05),but not from that in Beijing Han and Africans(P>0.05).(4)Three genotypes AG,AA and GG were detected at rs4968168 of SOCS3 gene in the asthma group and healthy group of Heiyi Zhuang nationality.Two alleles were A and G.The genotype frequency in the asthma group was AG 39.0%,AA 29.0% and GG 32.0%.The allele frequencies of A and G were 48.5% and 51.5%.The genotype frequency in the healthy group was AG 48.0%,AA14.0% and GG 38.0%.The allele frequencies of A and G were 43.5% and 56.5%.There were statistic discrepancies in gene frequency and allelic frequency at this locus between the two groups(P<0.05).Compared with GG genotype,the risk of asthma in AA genotype was2.460 times higher(OR=2.460,95%CI=1.114-5.433,P=0.019),the risk of asthma of A allele was 1.537 times higher than that of the G allele(OR=1.537,95%CI=1.032-2.288,P=0.035).Comparison of the data in the rs4969168 locus with the ensemble database yielded that the genotype frequency of this locus in the Heiyi Zhuang population was significantly different from that in Americans,Europeans,Africans and South Asians(P<0.05),but not from that in Beijing Han(P>0.05).The allele frequency of this locus in the Heiyi Zhuang population was significantly different from that in Beijing Han,Americans,Europeans,Africans and South Asians(P < 0.05).(5)Three genotypes AG,AA and GG were detected at rs4968170 of SOCS3 gene in the asthma group of Heiyi Zhuang nationality.Two alleles were A and G.The genotype frequency in the asthma group was AG 8.0%,AA 90.0% and GG 2.0%.The allele frequencies of A and G were 94.0% and 6.0%.Two genotypes AG and AA were detected at rs4968170 of SOCS3 gene in the asthma group of Heiyi Zhuang nationality.Two alleles were A and G.The genotype frequency in the healthy group was AG13.0% and AA 87.0%.The allele frequencies of A and G were 93.5% and 6.5%.There was no statistic discrepancy in gene frequency and allelic frequency at this locus between the two groups(P > 0.05).Comparison of the data in the rs4969170 locus with the ensemble database yielded that the genotype frequency and the allele frequency of this locus in the Heiyi Zhuang population was significantly different from that in Americans,Europeans,Africans and South Asians(P <0.05),but not from that in Beijing Han(P > 0.05),with G alleles being low-frequency alleles.Conclusion:(1)SNPs at rs4969168 in the SOCS3 gene was associated with the incidence of asthma in children of Guangxi Heiyi Zhuang nationality and the AA genotype is susceptible genotype.SNPs at rs9914220 and rs4969170 in the SOCS3 gene were not associated with the incidence of asthma in children of Guangxi Heiyi Zhuang nationality.(2)The genotype and allele frequencies of rs9914220,rs4969168 and rs4969170 in the SOCS3 gene are regionally different between Heiyi Zhuang and some other ethnic groups.
