| Background : Colorectal cancer(CRC)is a kind of malignant tumor with high incidence in the world,which poses a serious threat to human life and health.It has become an important research to find more safe and effective anticancer substances from natural plants.Ginger Exosome-like Nanovesicles(GELN)are nanoparticles that contain lipids and proteins,nucleic acids,metabolites and other active substances.It has shown a certain therapeutic effect on intestinal diseases and has a certain therapeutic potential for colorectal cancer,but its anti-Colorectal cancer effect and mechanism is still unclear.Objective: To investigate the effect and mechanism of ginger exosome-like nanovesicles in colorectal cancerMethods :(1)The ginger exosome-like nanovesicles were extracted by PEG polymer precipitation and ultracentrifugation combined with sucrose density gradient centrifugation;(2)Trypan blue staining,colony formation,Ed U and Annexin V-FITC/PI double staining were used to detect the effects of ginger exosome-like nanovesicles on the cell viability,proliferation and apoptosis of CRC cells;(3)Tumors were implanted subcutaneously on the nude mice and treated with ginger exosome-like nanovesicles,tumor size was observed,IHC were used to detect the expression of Ki-67 in cancer tissues,TUNEL immunofluorescence were used to detect the apoptosis in tumor tissues;(4)Proteomics was used to detect the differential protein expression of CRC cells treated with ginger exosome-like nanovesicles.Bioinformatics analysis was conducted to explore the anti-colorectal cancer mechanism of ginger exosome.Western blot was used to verify the proteomics results.Results :(1)Ultracentrifugation combined with sucrose density gradient centrifugation and PEG polymer coprecipitation can successfully extract ginger exosome-like nanovesicles,and the size of ginger exosome-like nanovesicles is about 100 nm.(2)Ginger exosomelike nanovesicles can inhibit cell viability and proliferation of colorectal cancer cells,induce apoptosis of colorectal cancer cells,inhibit tumorforming ability of colorectal cancer cells in vivo,and induce apoptosis of CRC cells in nude mouse tumor tissue.(3)Proteomic results showed that ginger exosome-like nanovesicles could induce apoptosis of CRC cells via mitochondrial pathway,and differential proteins were enriched in PI3K-Akt pathway.Conclusion: Ginger exosome-like nanovesicles can inhibit the proliferation of colorectal cancer cells and induce the apoptosis of colorectal cancer cells through mitochondrial apoptosis pathway. |
