| Rosacea is a chronic,relapsing inflammatory skin disease mainly affecting the face,It is characterized by recurrent episodes of flushing,persistent erythema,papules,pustules,skin thickening and enlargement.Abnormal activation of immune responses and vascular dysfunction are the mechanism of rosacea.Aspirin,one of the nonsteroidal anti-inflammatory drugs(NSAIDs),is widely used for numerous inflammatory diseases because of its multiple pharma cological properties,such as anti-inflammatory,inhibit immune response and angiogenesis.Therefore,we hypothesized that aspirin might have a curative effect on the treatment of rosacea。Objective:To investigate the possible effect and mechanism of aspirin in the treatment of rosaceaMethods:BALB/c female mice were randomly divided into model group(solvent+LL37),blank control group(solvent+PBS),aspirin treatment group(aspirin+LL37)and aspirin control group(aspirin+PBS).Mice in model group were intraperitoneally injected with solvent for 4 days every day,and on the 3rd and 4th day,LL37 was injected into the dorsal skin at the same time for mouse model of rosacea.The blank control group was given intraperitoneal injection of solvent for 4 days,and on the 3rd and 4th day,intradermal injection of PBS on the dorsal skin was performed simultaneously.In the aspirin treatment group,aspirin solution was intraperitoneally injected for 4 days,and LL37 intradermal injection was performed on the back skin at the same time on the 3rd and 4th day.In the aspirin control group,aspirin solution was intraperitoneally injected for 4 days,and on the 3rd and 4th days,PBS was intradermally injected into the back skin.12h after final injection,the skin appearance of the back of each group of mice was photographed and recorded.Skin tissue samples were collected from each group,and the histopathological changes of mouse skin were observed by hematoxylin-eosin staining(HE staining).Application of real-time fluorescent quantitative of nucleotide polymer chain reaction(Real-timeQuantitativepolymerase chain reaction,RT-qPCR)technology to detect each group of chemokines,cytokines and other expression level,immunofluorescence test groups of CD4+ T cells in skin tissue,The number of CD31 positive blood vessels.Result:1.Aspirin ameliorates the phenotype and inflammatory cell infiltration in the dermis of LL37-induced rosacea-like mice model.2.Aspirin decreases the expression of inflammatory cytokines(Il-6,Tnf-α)and chemokines(Ccl3,Ccl5,Ccl20,Cxcl10,Cxcl11,Cxcl12 和Cxcl15)associated with rosacea.3.Aspirin represses the CD4+ T cell infiltration,markedly reduce the expression of Thl-associated gene(Stat1,Ccr5)and Th17-associated gene(Stat3,Il-20,Ccr6和I1-17)in LL37-induced rosacea-like mice.4.Aspirin inhibits angiogenesis in the skin(CD31)and the expression of Vegf of LL37-induced rosacea-like mice.Conclusion:Aspirin ameliorates rosacea phenotype by inhibiting the immune response and angiogenesis.It can be a promising drug for the treatment of rosacea. |
