Objective:Osteosarcoma,a malignant bone tumor represented by U2 OS cell line.,can be treated by surgical excision.In addition to the level of tumor growth,recurrence and metastasis has gradually attracted the attention of researchers.Propofol is the intravenous anesthetics,.Recently,studied has shown that propofol can affect the movement of cancer cells in vivo and in vitro,promoting reproduction and metastasis of cancer cells.Therefore,propofol may be a better clinical anesthetic than other drugs.Studies have shown that AQP1 is highly expressed in osteosarcoma cells and can participate in the development of osteosarcoma cells,However,it is unclear whether propofol affects the biological activity of osteosarcoma by regulating AQP1 expression.In this study,the effects of propofol on osteosarcoma cells and the expression changes of AQP1 were clarified through the incubation of osteosarcoma cells with different concentrations of propofol,and then transfection reagents were added to further verify whether propofol affects the expression of osteosarcoma cells through AQP1.Method:1、Osteosarcoma cell culture and immunofluorescence staining were performed.2、Osteosarcoma cells were incubated with different concentrations of propofol,and the effects of propofol on the activity and migration ability of osteosarcoma cells were verified by CCK8 cell activity test and scratch test.3、The expression level of AQP1 was detected by PCR and Western blot.4 、Transfection reagent was added to make AQP1 overexpression in osteosarcoma cells.Blank control group,special ineffective transfection AQP1 group,special transfection AQP1 group,propofol + ineffective transfection AQP1 group,propofol + transfection APQ1 group were set,respectively.The expression level of AQP1 in transfected osteosarcoma cells was detected by PCR and Western blot,and scratch test was used to confirm whether propofol regulates the migration ability of osteosarcoma cells through AQP1.Result: 1、AQP1 is highly expressed in osteosarcoma cells.2、CCK8 test showed that the activity of osteosarcoma cells’ activity decreased with increasing propofol concentration.The scratch test showed that the cell migration ability decreased gradually with increasing propofol concentration.3、PCR and Western blot showed that with the increase of propofol concentration,AQP1 expression decreased.4、After transfection,PCR and Western blot showed that AQP1 expression was increased in the AQP1 transfection group.There was no significant difference between the ineffective transfection group and the blank control group,while the expression of AQP1 was significantly inhibited in the transfection group with propofol incubation.5、Scratches after transfection showed that cell mobility increased in AQP1 transfection group alone.There was no significant difference between the ineffective transfection group and the blank control group.Propofol incubation in AQP1 transfected group significantly decreased cell mobility.Conclusion:1、The activity and migration of osteosarcoma cells were inhibited by propofol.2、Propofol inhibits AQP1 expression in osteosarcoma cells and inhibits its activity and migration ability...
