Study On An Injectable Adhesive Hydrogel With Antibacterial And Promoting Cell Growth For Skin Wound Healing

Research purpose:This study intended to explore a new type of injectable and low-cost hydrogel dr essing with certain adhesive and simple synthesis method,which can resist to bacteria and prompt wound healing.By adjusting different synthesis ratios,we investigated th e feasibility of preparing this hydrogel as a wound dressing,and explored the applicati on prospect of the hydrogel synthesized by the optimal proportion based on the releva nt characterization.Research method:In this paper,a novel injectable hydrogel(CPH)wound dressing was constructed based on carboxymethyl chitosan(CMCS)and polyethyleneimine(PEI).The two can physically interact through strong hydrogen bonds.A series of hydrogels(CPH)were prepared by mixing 5wt%CMCS aqueous solution and 5wt%PEI aqueous solution a t the volume ratio of 7:3,5:5 and 3:7,respectively.The structure was characterized by Fourier transform infrared(FTIR),scanning electron microscopy(SEM),Zeta potential analysis,and other means.Through swelling rate detection,adhesion,detection,injecta bility,water impact resistance,rheological and antibacterial performance,in vitro bioco mpatibility testing and in vivo animal experiments(HE staining and Masson staining)and other characterizations,it was evaluated that the new hydrogel has a good applic ation prospect as a wound dressing.Research result:In this study,we successfully prepared CPH hydrogel dressings with different pr oportions.The synthesis method of the hydrogel is simple.By characterizing its basic properties,we found that the hydrogel has a stable gel structure,certain pore structur e and adhesion properties(7/3CPH group:0.51k Pa,5/5CPH group:0.71k Pa,3/7CPH group:0.87k Pa),good water absorption,injectability and water carrying impact capac ity.Then,by characterizing the biocompatibility of the materials and conducting MTT and live death staining experiments,we found that the cell survival rates of the 7/3CP H group,5/5CPH group,and 3/7CPH group on day 7 were 102.3%,104.1%,and 19.8%,respectively,and the former two have a certain ability to promote cell growth.Th e hemolysis rate test results of the three groups showed that the hemolysis rate of bloo d cells was lower than the evaluation standard of 5%,which indicated that the dressin g had a low effect on the hemolysis of red blood cells and met the relevant standards of biological materials.We further investigated the antibacterial performance of the m aterial and found that the the CFU of Staphylococcus aureus in the blank group,7/3C PH group,5/5CPH group,and 3/7CPH group were 56.7×10~7CFU/m L,8.9×10~7CFU/m L,2.5×10~7CFU/m L,and 0 CFU/m L,respectively,and the CFU of Escherichia coli i n the blank group,7/3CPH group,5/5CPH group,and 3/7CPH group were 66.0×10~7C FU/m L,30.9×10~7CFU/m L,19.0×10~7CFU/m L,and 8.5×10~7CFU/m L,respectively.The three groups of CPH showed good antibacterial properties.In addiction,bacterial ele ctron microscopy experiments also reflected the antibacterial effect of the material fro m the microscopic level.Finally,animal experiments in rats showed that the healing r ate of the blank control group was 88.48%,that of the 7/3CPH group was 94.58%,tha t of the 5/5CPH group was 98.02%,and that of the 3/7CPH group was 93.16%.Mater ials as dressings have an impact on wound healing.HE and Masson experiments on ti ssue sections further showed that the 7/3CPH and 5/5CPH hydrogel treatment groups had the thickest and most obvious collagen deposition in wound granulation tissue.Conclusions:The CPH hydrogel prepared with CMCS solution and PEI solution at a volume r atio of 5:5 has the most comprehensive properties.The hydrogel is easy to prepare,in jectable and can resist water flow impact,with certain adhesive,pore structure,and g ood biocompatibility,which can resist to bacteria and promote wound healing.Chitos an has a wide range of natural sources and is relatively low in cost.The preparation m ethod of the hydrogel is simple without complex chemical synthesis.Therefore,the C PH hydrogel has a good application prospect.
