Salidroside is one of the main active components of traditional Tibetan medicine Rhodiola Rosea,which has many pharmacological activities such as anti-fatigue and anti-oxidation.Salidroside is mainly extracted from rhodiola rosea,a natural plant,but its poor growth environment and slow growth are important factors limiting its reserves.Salidroside has many pharmacological activities.In view of its strong development potential,it is particularly important to explore other sources of salidroside.Among them,chemical synthesis is an effective way.At present,researches on the total synthesis of salidroside mainly focus on pharmacology and synthesis,and there are few reports on the quality of the total synthesis of salidroside.Therefore,this paper conducted a comprehensive quality study on the total synthesis of salidroside.In this paper,a sample of salidroside was made by referring to relevant literature,and the quality of synthetic salidroside samples was analyzed deeply.Mass spectrometry,hydrogen NMR spectroscopy,carbon spectroscopy and infrared spectroscopy were used to confirm the structure of the compounds,their physical and chemical properties were determined,and identification tests were carried out by chemical identification,spectral identification and chromatographic identification.Based on the synthetic route,a method for the detection of related substances of total synthetic salidroside was established,and the structure of process impurities and degradation impurities was confirmed and speculated.It is speculated that impurity a may be 4-(2-hydroxyethyl)phenylβ-D-glucoside,impurity c may be 4-[2-(β-Dglucopyranyl)ethyl]-4-hydroxy-2,5-cyclohexadiene-1-one,and impurity b is determined to be tyrosol and impurity D is p-hydroxybenzaldehyde,providing reference for subsequent quality research.The detection method of residual solvent was established to detect petroleum ether,acetone,ethyl acetate,methylene chloride,methanol and anhydrous ethanol using the same method.In this paper,the content of synthetic salidroside was determined by HPLC.Based on the experimental results,a draft quality standard for synthetic salidroside was established.
