| Objective: To study the compound components,key targets and multi-path mechanism of Shenfu injection in the treatment of cardiac arrest(CA).To provide theoretical reference for further study of Shenfu injection.Methods: The main chemical components of Shenfu injection were found out from the laboratory data provided by the relevant literature of CNKI,Wanfang Medicine and Pub Med database.The molecular structure of Shenfu injection compounds was obtained by searching Pubchem,and submitted to the pharmapper integrated pharmacophore matching platform to collect the target of compounds.The human gene database(Gene Cards)was used to predict the disease target.Use the function of the "Venn Diagram" installation package of R software to intersect the drug target and the disease target to obtain a common target,and upload the obtained target to STRING database for protein-protein interaction analysis.The drug-disease-component-target network was constructed by using Cytascape software,and GO function enrichment analysis and KEGG pathway analysis were performed in omicshare database.Results: A total of 28 chemical components of Shenfu injection were collected,including 374 co-action targets related to cardiac arrest.The core targets were CTNNB1(catenin beta 1)、NOTCH1(notch receptor 1)、ERBB2(erb-b2 receptor tyrosine kinase 2)、ESR1(estrogen receptor 1)、CDC42(cell division cycle 42)、VEGFA(vascular endothelial growth factor A),etc.The GO biological process results showed that most of the genes belonged to the intracellular part,and the function of the daughter was mainly related to protein binding;Biological processes are mainly embodied in metabolic processes.The results of enrichment analysis of KEGG pathway showed that it mainly involved MAPK signaling pathway、HIF-1 signaling pathway、Rap1 signaling pathway、Ras signaling pathway、VEGF signaling pathway,etc.Conclusion: This study preliminarily identified the main target and pathway of Shenfu Injection in the treatment of cardiac arrest from the perspective of network pharmacology,providing some reference for further research... |
