Objective: In order to explore the medication rule of traditional Chinese medicine in the treatment of hyperprolactinemia,the published literature on the treatment of hyperprolactinemia was analyzed by data mining technology.analyze the drug rules for the treatment of hyperprolactinemia and make a network pharmacological analysis of the core drugs obtained from data mining,so as to explore its possible mechanism,so as to provide reference for the clinical treatment of hyperprolactinemia.Methods: The literatures on the treatment of hyperprolactinemia by traditional Chinese medicine published in China knowledge Network Database,VIP Database and Wanfang Database from 1992.10.01 to2022.10.01 were reviewed.After the inclusion and exclusion of the criteria into the EXCEL,and standardized processing of the data to build a drug prescription database;hyperprolactinemia syndrome statistics and prescription statistics,the frequency,types,four qi,five flavors and meridian of traditional Chinese medicine were analyzed,and then the statistical software IBMSPSSModeler18.0 and IBMSPSSStatistics21.0 were used for drug association rules,drug cluster analysis and so on.The core drug combinations were obtained according to drug frequency and association rules,then the active components of core drugs were screened by TCMSP database,and their targets were predicted.The related targets of hyperprolactinemia were selected by GeneCards and OMIM databases."traditional Chinese medicine-component-target network map" and "hyperprolactinemia-related components-target-network map" were built by Cytoscape3.8.0 software to find the key active components of core drugs in the treatment of hyperprolactinemia.The drug-disease protein interaction network diagram(PPI)was generated by String platform,and then introduced into Cytoscape3.8.0 for topological analysis to find the key targets of core drugs in the treatment of hyperprolactinemia.Use cluster Profiler R package for running analysis,and get the results of GO and KEGG analysis.Results: In terms of data mining,84 literatures were included in this study,and 95 prescriptions were extracted.The main TCM syndromes of hyperprolactin were liver depression.In addition to the self-designed formula,Xiaoyao powder is the most commonly used addition and subtraction.Top 5high frequency drugs were Paeony,malt,bupleurum bupleurum,angelica sinensis and licorice.Among the drugs,the frequency of use of warm drugs was the highest,followed by flat drugs and cold drugs.The main medicinal tastes are sweet,bitter and xin.The main channel into the liver,spleen,kidney three channels.26 combinations of two drugs,67 combinations of three drugs and 45 combinations of four drugs were obtained by association rule analysis,and the core drug combinations of angelica sinensis,bupleurum bupleurum,paeony and malt for the treatment of hyperprolactin were obtained.Four prescriptions were obtained by cluster analysis.In terms of network pharmacology,45 active ingredients of core drugs were screened out through TCMSP database,134 active ingredient targets were obtained after weight removal,202 disease targets related to hyperprolactinemia were obtained from GeneCard and OMIM disease databases,and 26 common targets of core drugs and diseases were obtained by intersection of the two.By introducing Cytoscape3.8.0 software to build the "TCM-Component-target network map",the key components of core drugs in the treatment of hyperprolactinemia were screened according to the Degree value,and the main active components such as kernelol,luteolone and paeoniflorin were identified.The PPI network map was established and topological analysis was carried out.It was concluded that the key targets for the treatment of hyperprolactinemia with core drugs were AKT1,IL6,EGFR,etc.GO and KEGG enrichment showed that the potential targets of core drugs in the treatment of hyperprolactinemia were mainly distributed in the components of synaptic membrane,the intrinsic components of synaptic membrane and the base of cells.Its molecular functions are mainly related to the activity of cytokines and growth factors.Biological processes mainly include glandular development,the activity of positive regulatory protein serine threonine kinase,and the regulation of cell-cell adhesion.KEGG obtained 187 results including pancreatic cancer,JAK-STAT signaling pathway,FoxO signaling pathway and other pathways.Conclusion: The main TCM syndrome types of hyperprolactinemia are liver depression.The core drugs angelica,bupleurum bupleurum,Radix paeoniae alba and malt play a therapeutic role by playing a multi-component,multi-target and multi-pathway regulation target of hyperprolactinemia.The main mechanism may be related to inhibiting cell growth,regulating cell differentiation,promoting cell apoptosis and regulating growth and development. |
