| Objective:The therapeutic effects of resveratrol and platelet-rich plasma on cartilage injury in rats with knee osteoarthritis were observed and discussedMethod:1.Experimental animal groups: a total of 30 SD male rats were randomly divided into 5 groups: sham operation group(1)(control group),knee osteoarthritis model grou(2);resveratrol intervention group(3);PRP intervention group(4)and resveratrol plus PRP intervention group(5).2.Experimental animal modeling and intervention measures: the improved Hulth method was used to establish the animal model of knee osteoarthritis.After the completion of the modeling,the sham operation group only opened the knee joint;the model group performed the modeling operation on the rats in the group;the resveratrol intervention group was given intragastric administration of resveratrol every day;and the PRP intervention group : after the modeling was completed,the knee joint on the modeling side was injected with PRP every week;Resveratrol combined with PRP intervention group : after the completion of modeling,rats were given resveratrol intragastric administration combined with weekly knee joint PRP injection.3.24 hours after the last administration,the rats were killed by cervical dislocation method to observe the knee joint cartilage of each group;the blood was taken from abdominal cavity to detect the content of IL-1 β,IL-6 and TNF-α in serum by Elisa method;the cartilage repair was observed by HE staining microscope after paraffin sections of knee cartilage tissue were made;the apoptosis of chondrocytes in each group was detected by Tunel method.The protein expressions of Sirt1,Coll Ⅱ and MMP-13 in knee joint chondrocytes of rats in each group were detected by Western Blot method.Results:The results of HE staining showed that resveratrol and PRP had obvious therapeutic effects on articular cartilage injury caused by OA,and the changes under microscope were better than those in the model group;at the same time,the therapeutic effect of resveratrol combined with PRP intervention group was better than that of the two treatments alone;compared with the microscopic changes of resveratrol intervention group and PRP intervention group,the experimental results temporarily supported that the therapeutic effect of resveratrol was better than that of PRP.The results of Elisa showed that the levels of serum IL-1 β,IL-6 and TNF-α in resveratrol intervention group,PRP intervention group and resveratrol plus PRP intervention group were significantly lower than those in the model group(P < 0 01),suggesting that the intervention measures adopted in this experiment successfully reduced the inflammatory severity of osteoarthritis in rats and had anti-inflammatory effects on the increase of inflammatory factors caused by OA.However,there was no significant difference in the levels of IL-1 β,IL-6 and TNF-α in serum of rats.Tunel staining showed that the apoptosis rate of chondrocytes in the model group was significantly higher than that in the sham operation group,suggesting that osteoarthritis could cause chondrocyte apoptosis,while the apoptosis rate of chondrocytes in the resveratrol,PRP and combined intervention group was significantly lower than that in the model group,suggesting that resveratrol and PRP could inhibit chondrocyte apoptosis or promote chondrocyte proliferation.The apoptosis rate of resveratrol combined with PRP group was significantly lower than that of resveratrol plus PRP group,and the difference was statistically significant(P < 0.05).It is suggested that resveratrol combined with PRP can effectively inhibit chondrocyte apoptosis or promote its proliferation,and the combination of resveratrol and PRP has more obvious effect on inhibiting chondrocyte apoptosis than that of resveratrol alone and may have synergistic effect.There was no significant difference between resveratrol intervention group and PRP intervention group.The results of Westernblot showed that the expression of Sirt1 in resveratrol intervention group,PRP intervention group and resveratrol plus PRP intervention group was significantly higher than that in model group and sham operation group.However,there was no significant difference in Sirt1 expression among resveratrol intervention group、PRP intervention group and resveratrol plus PRP intervention group.The expression of Coll Ⅱ protein in the model group was significantly lower than that in the other four groups,which suggested that the injury and destruction of chondrocytes were very important in the course of osteoarthritis,while the expression of Coll Ⅱ in resveratrol intervention group,PRP intervention group and resveratrol plus PRP intervention group was higher than that in model group.The expression of MMP-13 in the model group was significantly higher than that in the other four groups.Compared with the model group,it was found that the degradation of Coll Ⅱ by MMP-13 decreased significantly after the intervention of resveratrol and PRP.However,the comparison of the expression of MMP-13 among the resveratrol intervention group、PRP intervention group and resveratrol plus PRP intervention group,did not support the superposition or synergistic therapeutic effect of the resveratrol and PRP.Conclusion:1.Resveratrol and platelet-rich plasma play a good role in the treatment of cartilage injury in rats with knee arthritis;resveratrol,PRP intra-articular injection and their combination can be used in the treatment of osteoarthritis,and can slow down the injury of articular cartilage,promote the proliferation of chondrocytes and the recovery of cartilage tissue.2.The effect of the combination of resveratrol and PRP does not support the synergism and superposition of resveratrol and PRP.3.Resveratrol and PRP have some common ways to play the role of antiinflammation and anti-apoptosis in the treatment of osteoarthritis. |
