| Background: Malignant tumor has become the main cause of death of most patients.China is a country with a high incidence of gastric and colorectal cancer,and its morbidity and mortality are in the forefront of tumors in our country.At this stage,surgical resection is still the main treatment for malignant tumors,supplemented by radiotherapy,chemotherapy and targeted immunotherapy.Xihuang Pills,as a traditional medical treasure in China,is also a famous anti-tumor prescription in China,which has an inhibitory effect on a variety of tumors.Objective: to study the mechanism and target of Xihuang Pills in anti-colorectal cancer based on network pharmacology,and to explore the mechanism of Xihuang Pills in promoting apoptosis of gastric cancer and colon cancer cells.Objective: Based on network pharmacology,to study the mechanism of Xihuang Pills on colorectal cancer and screen its target,and to explore the mechanism of Xihuang Pills in promoting apoptosis of gastric cancer and colon cancer cells.Method:1.The action targets of four main components of Xihuang Pills on colorectal cancer were screened by network pharmacology platform and bioinformation technology,and the potential signal pathways were screened out.2.The inhibitory effect of Xihuang Pills on the growth of human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells was studied by cell proliferation toxicity(CCK-8).3.Flow cytometry was used to detect the effect of Xihuang Pills on the arrest of cell cycle progression in human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells.4.Western blotting was used to verify the regulatory effect of Xihuang Pills on the expression of E2F1,Cyclin B1 and CDK1 proteins in human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells.Results:1.There are 264 common target genes of Xihuang Pills and colorectal cancer.IL1 B,IL6,TNF,ICAM1 and other targets may act on colorectal cancer and play an anti-tumor effect.These targets may also play a role in the treatment of colorectal cancer through IL-17,TNF,AGE-RAGE,C-typelectinreceptor,PI3 K,MAPK and other signal pathways.The expression of CCND1,CCDB1,CXCL11 and other genes is related to the cell cycle and up-regulated in colorectal cancer tissue,and it improve that Xihuang Pills treatment of colorectal cancer may be related to the progression of cell cycle.2.The results of cell proliferation-toxicity(CCK-8)test showed that Xihuang Pills had a concentration-and time-dependent inhibitory effect on human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells.3.Xihuang Pills blocks the cell cycle transformation of human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells.4.Xihuang Pills down-regulated the expression of E2F1,Cyclin B1 and CDK1 protein,regulated cell cycle and promoted cell apoptosis.Conclusion: Network pharmacological analysis shows that Xihuang Pills has the common target of anti-tumor effect and colorectal cancer disease,which provides the basis of signal pathway for Xihuang Pills to promote the apoptosis of gastric cancer and colon cancer cells.From the aspects of cell and protein expression,it has been proved that Xihuang Pills can promote the apoptosis of human gastric adenocarcinoma AGS cells and human colon cancer SW480 cells by regulating the expression of E2F1,Cyclin B1 and CDK1.E2F1,Cyclin B1 and CDK1 are expected to be the important potential mechanisms of Xihuang Pills to exert its anti-tumor effect. |
