ObjectiveTo discuss the improvement effect of baicalin on myocardial hypertrophy and apoptosis induced by abdominal aorta ligation of the rats and to clarify the potential mechanism.MethodsThe model of cardiac hypertrophy was established by ligating abdominal aorta.The SD rats were randomly divided into control group,model group,low dose of baicalin(50 mg·kg-1)group and high dose of baicalin(100 mg·kg-1)group.After six weeks,the left ventricular ejection fraction(LVEF),left ventricular posterior wall end diastolic thickness(LVPWd),left ventricular posterior wall end systolic thickness(LVPWs),interventricular septal end diastolic thickness(IVSd)and ventricular septal end systolic thickness(IVSs)were measured by echocardiography;the heart weight indexes(HWI)and the weight of left ventricular index(LVWI)of the rats in various group were calculated;HE staining was used to observe the morphology of the myocardial cell of the rats in various group;the serum levels of interleukin-8(IL-8),interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)and interleukin-6(IL-6)of the rats in various group were detected by ELISA.The expressions of B-cell lymphoma 2(Bcl-2),Bcl-2 associated X protein(Bax),apoptosis-inducing factor(AIF)and calpain-1(calpain-1)protein in myocardial tissue of the rats in various group were detected by Western blotting.ResultsCompared with control group,LVEF was significantly decreased,LVPWd,LVPWs,IVSd,IVSs,HWI and LVWI were significantly increased(P<0.05),the serum levels of IL-8,IL-1β,TNF-α and IL-6 were increased(P<0.05),the myocardial cell were disordered arrangement,the expression levels of Bax,AIF and calpain-1 proteins in myocardial tissue of the rats in various group were increased(P<0.05),while the expression level of Bcl-2 protein was decreased(P<0.05),and the ratio of Bax/Bcl-2 was increased(P<0.05).Compared with model group,the LVEF of the rats in low and high doses of baicalin groups was increased,LVPWd,LVPWs,IVSd,IVSs,HWI and LVWI were decreased(P<0.05);the serum levels of IL-8,IL-1β,TNF-α and IL-6 were decreased(P<0.05);the myocardial cells were arranged in order,the expression levels of Bax,AIF and calpain-1 proteins in myocardial tissue of the rats in low and high doses of baicalin groups were decreased(P<0.05),while the expression level of Bcl-2 protein was increased(P<0.05),and the ratio of Bax/Bcl-2 was decreased(P<0.05).Compared with low dose of baicalin group the expression levels of HWI,IL-1β,TNF-α,IL-6 and Bax in high dose of baicalin group were decreased(P<0.05),but there was no statistical significance in other indexes(P>0.05).ConclusionsThe results indicated that baicalin could improve the myocardial hypertrophy,inflammation and apoptosis induced by abdominal aortic ligation in rats,and its mechanism might be related to the regulation of calpain-1/AIF signaling pathway.⑴ Baicalin can alleviate myocardial hypertrophy induced by abdominal aorta ligation in rats.⑵ Baicalin could alleviate the inflammatory response and myocardial cell apoptosis induced by abdominal aorta ligation in rats.⑶ Baicalin can inhibit the activation of calpain-1AIF signaling pathway in rats ligation through abdominal aorta.
