Introduction:Osteoporosis is a common clinical bone disease,often occurs in postmenopausal women and elderly patients.As the world’s population ages,the social and economic burden of osteoporosis is increasing.The characteristics of this disease are typical,often presented with a decrease in bone mass and deterioration of bone structure,thus increasing bone fragility and the risk of fracture.Clinically,patients are often treated for low back soreness,spinal deformation caused by vertebral compression fracture,and brittle fracture caused by non-traumatic or minor trauma.Despite significant advances in the understanding of the pathogenesis of osteoporosis and in the development of new drugs to treat osteoporosis,there are still challenges in giving appropriate treatment to patients who clearly need it.This is mainly due to concerns about the side effects and long-term effectiveness of treatment for osteoporosis patients.Therefore,there is an urgent need to improve patients’awareness of the advantages of anti-osteoporosis drugs and develop new drugs to treat osteoporosis.Objectives:By studying the correlation between long non-coding RNA ZBTB 40-IT 1,microRNA-514a-3 p/FOXO 4 axis and differentiation of human bone marrow mesenchymal stem cells,new ideas are provided for the clinical treatment of osteoporosis.Material and Methods:Human mesenchymal stem cells were isolated and cultured to establish a mouse model of OVX-induced osteoporosis.On this basis,the correlation between Lnc RNA ZBTB 40-IT 1,microRNA-514a-3 p/FOXO 4 axis and human bone marrow mesenchymal stem cell differentiation was verified through in vivo and in vitro experiments.Results:A series of experiments were conducted to verify the association between Lnc RNA ZBTB 40-IT 1,microRNA-514a-3 p/FOXO 4 axis and bone marrow mesenchymal stem cell differentiation.In summary,Lnc RNA ZBTB 40-IT 1 silences mediates differentiation of human mesenchymal stem cells to increase bone mass in osteoporosis conditions.Conclusions:Our study successfully demonstrated the regulatory role of long non-coding RNA ZBTB 40-IT 1 in the differentiation of human mesenchymal stem cells,and this molecular mechanism provides a new direction for the treatment of osteoporosis.As stem cell treatments have come to the fore in recent years,the current study may also provide evidence of this promising medical approach. |
