Objective: To study the effect and efficacy of the compound preparation of udabala-8flavor powder on pulmonary fibrosis and related factors.Methods: Forty C57BL/6 mice were randomly divided into blank group,blank + udabala-8miso group,model group,and udabala-8 miso group,10 mice according to body weight.Except for the blank group,the remaining two groups of mice were treated with bleomycin 5mg/kg,intratracheal injection molding,and administration was started on the 2nd day,distilled water was given in the blank group,CMC-Na gavage was given in the model group,and the udabala-8flavor group was dissolved with 0.5% CMC-Na to dissolve Udabara-8 flavor gavage,the volume of gavage was 20ml/kg,and after 28 days of administration,the mice were sacrificed,lung tissues were taken,and serum was used to detect relevant protein factors.Results: Lung coefficient: Compared with the model group,the lung coefficient of mice in the Udabala-8 powder group was significantly reduced,with a statistically significant difference(P<0.05).Histological observation: Compared with the model group,the damage of alveolar structure and the degree of alveolar atrophy in the lung tissue of the Udabala-8 powder group mice were significantly reduced,and the area of inflammatory cells and blue areas were significantly reduced.The difference was statistically significant(P<0.05);Immunohistochemical detection: Compared with the model group,the expression of type I collagen and type III collagen in the lung tissue of mice in the Udabala-8 powder group was significantly reduced,with a statistically significant difference(P<0.05).Protein Immunoblotting Assay for Protein Expression: TGF in Lung Tissue of Model Group Mice-β 1.The protein expression of a-SMA,smad2,and smad3 increased,and compared with the model group,the protein expression in the lung tissue of the Udabala-8 powder group mice decreased,with a statistically significant difference(P<0.05).The protein expression of smad6 and smad7 in the lung tissue of the model group mice decreased,and compared with the model group,the protein expression in the lung tissue of the Udabala-8 powder group mice was relatively increased,with a statistically significant difference(P<0.05).RT-QPCR method for detecting m RNA expression:TGF in lung tissue of model group mice-β 1.The m RNA expression of a-SMA,smad2,and smad3 increased;Compared with the model group,the expression level of Udabala-8 Weisan group decreased with statistical significance(P<0.05).The m RNA expression of smad6 and smad7 in the lung tissue of the model group mice decreased;Compared with the model group,the expression level of Udabala-8 Weisan group increased,and the difference was statistically significant(P<0.05).Conclusion: Udabara-8 flavor powder has a certain therapeutic effect on pulmonary fibrosis,and may inhibit the formation of fibrosis through TGF-β/Smad signaling pathway. |
