Abstract/Summary:
Objective:To investigate the effect of Zandan-3 on apoptosis by regulating the expression of Calumenin,the protein associated with mitochondria and apoptosis-associated proteins,so as to clarify the protective mechanism of ischemia reperfusion injury.Methods:Eighteen male SD rats were randomly divided into the control,MIRI,and Zandan-3 groups,or 10 rats per group.Rats in control and MIRI groups were infused with 2ml of 0.9% Naacl injection once daily;Zantan-3(0.3g/kg rats 1h before surgery)with 0.9% Naacl injection 2ml once a day.All rats were treated with gavage for 10 days.The model of MIRI was established between Zandan-3 and MIRI.After the model was successfully established,the myocardium was obtained for H&E staining,and the myocardial pathological changes were compared in different groups.The expression levels of Calumenin,DRP 1,Mfn 1,chop,Bax,Bcl-2 were measured,and the apoptotic rate of cardiomyocytes was measured by TUNEL method.Results:1.HE staining showed that myocardial damage was severe in the MIRI group compared with the control group,and less severe in the Zandan-3 group.2.According to the Western blot results,Compared with the control group,The expression of Calumenin,the mitochondrial fusion protein Mfn 1,and the anti-apoptotic protein Bcl-2 was significantly decreased in the MIRI group,However,the expression of mitochondrial fission protein DRP-1,apoptotic factor Chop and proapoptotic factor Bax increased significantly;In the Zanandan-3 group,The expression of Calumenin,mitochondrial fusion protein Mfn 1,and anti-apoptotic protein Bcl-2 was significantly higher than that in the MIRI group,The expression of the mitochondrial fission protein DRP-1,apoptotic factor Chop,and proapoptotic factor Bax was significantly lower than that in the MIRI group.3.TUNEL results showed that the apoptosis rate in MIRI group was significantly higher than control group,while the apoptosis rate in Zandan-3 group was significantly lower than that in MIRI group.Conclusion:1.Zandan-3 soup can effectively increase the expression of calcium binding protein(Calumenin)in cardiomyocytes,and then inhibit the expression of mitochondria-related protein DRP-1,increase the expression of mitochondrial fusion protein Mfn-1,reduce the expression of apoptosis-related proteins Chop and Bax protein,increase the expression of anti-apoptotic protein Bcl-2,significantly reduce the apoptosis and reduce the rate of apoptosis.2.Zandan-3 decoction reduces cardiomyocyte apoptosis through the Calumenin mitochondrial dynamics pathway,and subsequently protects ischemia-reperfusion cardiomyocytes.
