Abstract/Summary:
Objective:Through simulating drought stress of Artemisia argyi,the mechanism affecting the high quality and high yield of Artemisia argyi was clarified.Under the influence of extreme climate conditions,technical support was provided for further research on the optimization of cultivation conditions,drought resistance evaluation and the quality of medicinal materials.This study provides scientific basis for the cultivation of Artemisia annua in arid area.Methods:In this study,the drought stress of Artemisia annua was studied with different concentrations of PEG-6000 solution by pot method.Under the guidance of Li Hesheng,Principles and Techniques of Plant Physiological and Biochemical Experiments,physiological indexes such as biomass,chlorophyll,electrical conductivity,SOD,POD,MDA,soluble protein and soluble sugar were measured.The contents of chlorogenic acid,rutin and apigenin-7-O-β-D glucuronide in Artemisia SPP.Were determined by HPLC.Results:The experimental results showed that:(1)With the increase of PEG-6000 treatment concentration and time,Artemisia frigida root contribution rate,root/shoot ratio,stem/leaf ratio,MDA,soluble protein,soluble sugar and other indexes showed an obvious increasing trend.However,when the stress concentration reached 15%-20%,the leaf contribution rate and other physiological indexes were inhibited by different levels.(2)The contents of chlorogenic acid,rutin and apigenin-7-O-β-D glucuronide in Artemisia frigida were determined by HPLC.The results showed that the concentration of chlorogenic acid had a good linear relationship in the range of 0.162~3.24mg/m L(r=0.9997).The concentration of rutin had a good linear relationship in the range of 0.00256~0.0512mg/m L(r=0.9999).The concentration of apigenin-7-O-β-D glucuronide had a good linear relationship in the range of 0.404 to 3.636mg/m L(r=1).RSD of reproducibility,precision and stability tests were all less than 2%.The average recoveries of chlorogenic acid,rutin and apigenin-7-0-β-D were 102.57%,101.14% and 100.08%,respectively.RSDS were 0.4%,1.5% and 0.5%,respectively.Under different stress concentrations,the contents of chlorogenic acid and rutin in 5 samples were 5%,the highest in the fifth sample,which were higher than other samples.The highest content of apigenin-7-O-β-D glucuronide was found at 10% stress concentration,which was the highest at the fifth sampling.Conclusions:Excessive drought has an effect on the growth and development,yield and quality of Artemisia frigida,while moderate drought stress has no obvious effect on the biomass of Artemisia frigida.But it can promote the increase of active ingredient content.Therefore,attention should be paid to water control during cultivation.the purpose of this study is to provide some theoretical basis for the efficient cultivation of Artemisia frigida,and to provide some theoretical reference for the research on the influence of drought environment on the accumulation of effective components in Artemisia frigida.
