| Objective:Through the ovarian granulosa cell apoptosis-autophagy pathway PI3K/AKT(phosphinositide-3 kinase/Alpha serine/threonine protein kinase),To investigate the molecular mechanism of acupuncture intervention on ovarian protection in rats with premature ovarian failure(POF).Methods:Vaginal smears were observed,and 60 Specific pathogen free(SPF)female Wistar rats with 3 consecutive estrous cycles were randomly selected,of which 45 rats were successfully established and divided into three groups:The model control group(n=15),acupuncture group(n=15)and medication group(n=15).Another 10 rats without premature ovarian failure were used as blank control group.The model was established by intraperitoneal injection of cyclophosphamide.The dose of cyclophosphamide was 50mg/kg on the first day,and then 8mg/kg per day for 14 days.The vaginal smears were observed to determine the disturbance of estrus cycle.The model control group did not receive any intervention,the acupuncture group was treated with Guanyuan(CV 4)and Sanyinjiao(SP 6)according to the acupoint combination obtained from our previous study and etiological analysis for 14 days,and the medication group was treated with 0.0025mg/kg by gavage for 14 days.After the intervention,Estradiol(E2)and Follicle-stimulating hormone(FSH)were detected by Enzyme-linked immune sorbent assay(Elisa).The concentrations of FSH and LH were detected by WesternBlot and Real-time quantitative polymerase chain reaction(RT-q-PCR).The expression of PI3K and AKT protein and mRNA was analyzed by RT-q-PCR.Results:Compared with the model group,the contents of estrogen in the acupuncture group and the medication group were significantly increased,while the contents of FSH and LH were decreased,which were close to the normal group.The values of PI3K and AKT in the acupuncture group were close to those in the normal group.The differences among the groups were statistically significant(p<0.05).In conclusion,acupuncture has a therapeutic effect on premature ovarian failure,which can affect the secretion of ovarian related hormones,and it is related to the expression of PI3K/AKT related proteins in the apoptosis-autophagy pathway.Conclusion:The mechanism of acupuncture in preventing premature ovarian failure may be related to the apoptosis-autophagy pathway represented by PI3K/AKT.Acupuncture can improve the hormone secretion of rats with premature ovarian failure,affect the expression of PI3K/AKT in the apoptosis-autophagy pathway,thereby inhibiting the apoptosis of ovarian granulosa cells,and play a protective role in the treatment of premature ovarian failure at the molecular biological level.In the process of follicular atresia,PI3K/AKT induces primordial follicle development and affects ovarian reserve function through the effect of its downstream bioactive factors. |
