| Purpose: The incidence and mortality rate of lung adenocarcinoma remain high,and improving the prognosis of lung adenocarcinoma patients is the focus of tumor treatment.Research has found that SENP3 is associated with proliferation and metastasis of various tumors,thus exploring the role of SENP3 in lung adenocarcinoma may help to discover new pathways for cancer treatment.This study aims to explore the clinical significance of SENP3 expression in lung adenocarcinoma and its effect on ferroptosis.Methods:(1)The expression levels of SENP3 between lung adenocarcinoma and normal lung tissues were analyzed using bioinformatics,and verified using q PCR technology.The association between SENP3 expression level and patient prognosis and clinical pathological features was analyzed,and COX regression was used to evaluate the prognostic value of clinical characteristics and SENP3 expression level in lung adenocarcinoma patients.The association between SENP3 expression level and immune cell infiltration and immunotherapy response in lung adenocarcinoma was studied.(2)The association between SENP3 and ferroptosis driver genes was analyzed.An RSL3-induced ferroptosis model of lung adenocarcinoma was established,and a stable SENP3-knockdown lung adenocarcinoma cell line was constructed and induced for ferroptosis.Differences in cell activity,ferroptosis indicators,and related proteins between the SENP3-knockdown group and control group were detected.Results:(1)The expression level of SENP3 is higher in lung adenocarcinoma than in normal lung tissue.(2)Lung adenocarcinoma patients with high SENP3 expression have a poorer prognosis than those with low SENP3 expression.The expression level of SENP3 is associated with patient clinical characteristics and is an independent prognostic factor for lung adenocarcinoma patients.(3)The expression level of SENP3 is negatively correlated with immune cell infiltration and immunotherapy response in lung adenocarcinoma.(4)SENP3 is associated with multiple ferroptosis driver genes.(5)RSL3 can induce ferroptosis in lung adenocarcinoma cell lines and upregulate the expression level of SENP3.(5)Knockdown SENP3 enhances RSL3-induced ferroptosis in lung adenocarcinoma cells.(6)Knockdown SENP3 downregulates the protein expression levels of SLC7A11 and GPX4 in lung adenocarcinoma cells.Conclusion:(1)The expression level of SENP3 in lung adenocarcinoma is related to the prognosis,clinical characteristics,and immune infiltration of lung adenocarcinoma patients.(2)Knocking down SENP3 expression can promote ferroptosis of lung adenocarcinoma cells.(3)Knockdown SENP3 downregulates the protein expression levels of SLC7A11 and GPX4. |
