BackgroundCancer is an important public health problem worldwide.Prostate cancer(PCa)is one of the common malignant tumors of the urinary system in adult men.Tumor metastasis is a situation that patients and doctors often face in the process of diagnosis and treatment,and often affects the treatment of the disease.Therefore,it is very important to explore the types and mechanisms of tumor cell metastasis for the treatment of tumors.Glutaminase(GLS)is the rate-limiting enzyme of intracellular glutamine catabolism and an important enzyme in human metabolism.It is widely distributed in different tissues,and its expression is up-regulated in various human tumors.Matrix metalloproteinases(MMPs)are a family of proteolytic enzymes.The enzymes of this family are similar in molecular structure and depend on Zn2+ for their composition.The extracellular matrix(ECM)is its main substrate.It plays a key role in the stabilization of normal tissue structure.Several studies have shown that MMP-2 and MMP-9 play an important role in promoting tumor metastasis.PurposeTo study the expression of GLS1,MMP-2 and MMP-9 in prostate cancer tissue,to study the relationship between their expression levels and the clinicopathological characteristics of prostate cancer,and to analyze the expression of several proteins in the disease,to understand the expression relationship of these proteins in prostate cancer.MethodsThe histopathological specimens of 40 cases of prostate cancer were collected(40 cases of benign prostatic hyperplasia were used as the control group),and the expression of GLS1,MMP-2 and MMP-9 in prostate cancer tissue and benign prostatic hyperplasia tissue were detected by immunohistochemical staining,and the three proteins were analyzed.Whether there is a difference in the expression between prostate cancer tissue and benign prostatic hyperplasia tissue,analyze the relationship between the expression of these proteins and the clinicopathological characteristics of prostate cancer disease,and analyze the correlation between the expression of these proteins in the disease.ResultsThis study showed that the expressions of GLS1,MMP-2,and MMP-9 were upregulated in prostate cancer pathological specimens,and there were significant statistical differences between the prostate cancer group and the benign prostatic hyperplasia group(P<0.01).There were differences in the expressions of MMP-2 and MMP-9 in different TNM stages(P<0.05).The results of Spearman correlation analysis showed that the expressions of MMP-2 and MMP-9 in prostate cancer pathological specimens were positively correlated(r=0.432,P<0.01),and the expressions of MMP-2 and GLS1 in prostate cancer pathological specimens were positively correlated(r=0.410,P<0.01),MMP-9 was positively correlated with the expression of GLS1 in prostate cancer pathological specimens(r=0.365,P<0.05),Gleason score was associated with blood PSA(r=0.538,P<0.01)、TNM staging(r=0.597,P<0.01)、PSAD(r=0.636,P<0.01)were significantly positively correlated,TNM staging was associated with MMP-2(r=0.369,P<0.05)、MMP-9(r=0.429,P<0.01)was positively correlated,and the expression of GLS 1 was positively correlated with PSA(r=0.354,P<0.05)and PSAD(r=0.314,P<0.05).ConclusionThis study showed that GLS1,MMP-2,and MMP-9 were significantly expressed in prostate cancer tissue.Among them,MMP-2 and MMP-9 were related to the TNM stage of the tumor.The higher the stage,the higher the expression level.PSA levels were not significantly correlated.The expression of several proteins in prostate cancer cells is positively correlated.The high expression of GLS1 may promote the production of MMP-2 and MMP-9,and their joint action may promote the local and distant metastasis of prostate cancer.
