1 BackgroundSepsis is a disorder in which the hosts homeostatic inflammatory response to infection leads to life-threatening organ dysfunction.The mortality rate of sepsis is very high,which seriously endangers public health and causes great economic burden to family and society.So far,the mechanism of sepsis is not fully understood.Early identification and aggressive treatment of sepsis can prevent progression of the disease and the occurrence of adverse outcomes.Therefore,it is an important research direction to explore new strategies for the treatment of sepsis,promising improvements in patient outcomes and survival rate.2 ObjectiveTo investigate the effect of the deacetylase inhibitor in histone,Trichostatin A,on the injury and apoptosis of human microvascular endothelial cell in lipopolysaccharide-induced sepsis model.3 MethodsHMEC cells was used as the research cells to establish LPS-induced septic cell model,which were divided into three groups:Phosphate Buffer(PBS)control group,LPS(5 μg/ml)stimulated group,TSA(500 ng/ml)+LPS(5 μg/ml)group.The cells of each group were cultured for 24 hours to 48 hours,and the concentration of lactate dehydrogenase(LDH)in the culture supernatant was detected by enzyme-linked immunosorbent assay(ELISA).The apoptosis rate was detected by Annexin V-FTTC/PI staining.Student’s t test was used for comparison between two independent groups.4 ResultsThe LDH level in the cultured supernatant increased significantly after it was stimulated by LPS after 24 and 48th hours.Moreover,the LDH level decreased statistically significantly(all P<0.05)in the cells treated with TSA,the apoptosis rate of cells treated with LPS 24 hours later was significantly higher than that in the control group,and the apoptosis rate of cells in TSA treated group was significantly lower in the LPS stimulated group(all P<0.05),the apoptosis rate of cells in LPS group treated by LPS 48 hours later was higher than that in PBS control group(P<0.05).Compared with the experimental group,the apoptosis rate of TSA intervention group was not significantly decreased(P=0.084).5 ConclusionTrichostatin A may has the protective effect of reducing cell injury and apoptosis in sepsis and has a certain timeliness.
