Background and objectArnebia euchroma(Royle)Johnst.,an herbal medicinal plant belonging to the family Boraginaceae,as a traditional medicinal plant,it has a long history of being used in the folk to treat burns,scalds and bacterial infections.Pharmacological studies shown that the extracts from A.euchroma has biological activities,such as antiinflammatory,antitumor,antibacterial,antiviral and antioxidant.Phytochemical investigation found that the chemical constituents of A.euchroma are mainly naphthoquinones,monoterpene phenols and quinones,phenolic acids,among which naphthoquinones are the main active components of A.euchroma.Inflammation is a defensive response of the body to pathological stimuli involved in the occurrence of cancer,infection and autoimmune diseases.A.euchroma has been widely used in the treatment of various inflammatory diseases,in view of the remarkable biological activity and clinical performance of A.euchroma.The chemical constituents and antiinflammatory activity of A.euchroma were investigated in this project,hoping to further expand our knowledge of the chemical constituents and anti-inflammatory activity of A.euchroma.MethodsSilica gel column chromatography,thin-layer chromatography,gel column chromatography and semi-preparative liquid phase were used to isolate the ethyl acetate parts of A.euchroma,and the structure of the compounds were determined by UV,IR,HR-ESI-MS and NMR spectroscopy;The chemical constituents of lipid fractions isolated from A.euchroma was analyzed by GC-MS;Three inflammation models were constructed by injecting LPS into the yolk sac of zebrafish,transecting zebrafish tail and immersing zebrafish in copper sulfate,and the naphthoquinone dimer ZCZ-3 and lipid fractions ZC-3,ZC-13,ZC-23 were screened for anti-inflammatory activity.ResultsTwelve compounds were isolated and identified,which were shikometabolin H(ZCZ-1),shikometabolin B(ZCZ-2),shikometabolin E(ZCZ-3),epoxyarnebinol(ZCZ-4),clavilactone A(ZCZ-5),arnebinol(ZCZ-6),2,3-secodiplopterol dioic acid(ZCZ-7),roburic acid(ZCZ-8),ergosta-4,6,8(14),22-tetraen-3-one(ZCZ-9),23ahomostigmast-5-en-3β-ol(ZCZ-10),24-ketocholesterol(ZCZ-11)and 24Ssaringosterol(ZCZ-12),where ZCZ-1,ZCZ-4 and ZCZ-7 were new compounds.A total of 15 fatty acids,6 steroids and 4 terpenoids were identified from ZC-3,ZC-13 and ZC-23 by GC-MS.The anti-inflammatory activity screening showed that ZCZ-3 and the lipid fractions ZC-3,ZC-13,ZC-23 had significant anti-inflammatory activity in a dose-dependent manner.ConclusionTwelve compounds were isolated from A.euchroma,and activity screening showed that the ZCZ-3 and lipid fractions isolated from A.euchroma had antiinflammatory activity,which further elucidation of the material basis of the medicinal effects of A.euchroma.
