| Objective:This article aims to study compared xining region(han)gastric cancer(Gastic Carcinoma,GC)the intestinal flora of difference between patients and healthy people.The significance of this study not only reflects that it has further enriched the relevant data of intestinal flora in Xining area for China’s intestinal flora database,but also explored the causes of high incidence of gastric cancer in Xining area,and provided some treatment methods in intestinal microecology for patients with gastric cancer in Xining area in the future,At the same time,it provides a new method and idea for the prevention of gastric cancer patients in Xining area by using landmark or different bacteria genus.Therefore,a thorough understanding of the etiology pathological classification and molecular level of intestinal flora of gastric cancer has laid a certain foundation for the prevention and treatment of gastric cancer patients in Xining area in the future。Methods: from 2020,the general clinical data and fecal samples of gastric cancer patients and healthy people who met the inclusion criteria in Qinghai Provincial People’s Hospital(the source of research objects is stable and representative)and met the inclusion criteria were collected(collected without any treatment and intervention).There were two groups in this study: Gastric Cancer Group(general clinical data and fecal samples)and healthy control group(general clinical data and fecal samples).DNA was extracted from fecal samples,and the 16 s-intestinal flora RNA sequencing data analysis.Results: 1.When the length of the sequencing sequence of the sample is between 301 and 400,the number of sequences measured is the most(close to800000).2.Through sample flora diversity fares(Estimators table)analysis found that gastric cancer group of intestinal microflora Simpson index is less than the control group(0.14262 vs 0.16032667,P < 0.05).So the intestinal flora in patients with gastric cancer group abundance than healthy controls(Control Check,CK);Although the gastric cancer group of intestinal microflora Shannon index greater than healthy controls,it was not statistically significant(P > 0.05).3.According to the rarefaction curve and Shannon Wiener curve,the richness of flora in gastric cancer group was higher than that in normal group;When the amount of sequencing data of the two groups is close to 30000,the curve tends to be flat,indicating that the amount of sequencing data is reasonable(more species cannot be detected by continuing to increase the sequencing depth).4.At the genus level,compared with phylum level and family level,the abundance of microbial communities in the two groups of samples is higher.Compared with the healthy control group,the proportion of lactic acid bacteria,antigen Streptococcus,Achromobacter,adlercreutiza,anaerostipes and anaerotruncus in gastric cancer group increased significantly,while the proportion of Bacteroides,the proportions of actinomycetes and acinetobacter were significantly reduced.5.At the gate level,the proportion of Firmicutes,cyanobacteria and Proteobacteria in the intestinal tract of patients with gastric cancer increased significantly;Bacteroidetes and actinomycetes significantly reduce the proportion of the door.6.At the family level,compared with the healthy control group,the proportion of Alphaproteobacteria,the number of clostridiaceae and clostridiaceae were significantly increased in gastric cancer patients.The proportion of actinomycetes,alcaligenaceae,acidaminococcaceae and aeromonadaceae decreased significantlycancer was significantly increased;The proportion of Actinobacteria,alcaligenaceae,acidaminococcaceae and aeromonadaceae decreased significantly.Conclusion: 1.Xining area of gastric cancer patients with intestinal flora diversity(Han nationality)is increased compared with the healthy control group.2.Xining region(han)gastric cancer group and health control group structure of intestinal flora difference was statistically significant. |
