Based On Data Mining And Network Pharmacology To Explore The Medication Rules Of Professor Mi Ziliang In The Treatment Of Chronic Atrophic Gastritis And The Mechanism Of Action Of His Core Prescriptions

Research purposes: Using data mining methods,to explore the medication rules of Professor Mi Ziliang in the treatment of chronic atrophic gastritis,and to use the network pharmacology method for Milao’s core prescription for the treatment of CAG to explore its mechanism of action in the treatment of CAG.Research methods: First,collect and organize the handwritten medical records of Professor Mi Ziliang’s treatment of CAG from 2016 to 2019,standardize the data and establish a database of medical records and prescriptions,and use statistical software to make statistics on the frequency of drugs used,properties and tastes,association rules,etc.At the same time,through data mining modules such as complex network analysis,the medication rules of Milao in the treatment of CAG were analyzed,and combined with the experience of teachers,the drug experience and academic thoughts of Milao were further summarized.On this basis,use TCMSP,HERB and Swiss Target databases to screen the active ingredients and action targets of drugs in core prescriptions,and obtain CAG disease targets from CTD,Gene Cards,OMIM,TTD databases,and use Venn platform to obtain the intersection of drugs and diseases,use Cytoscape3.9.0 to build drug-target-disease relationship network,use String database to build PPI network,carry out GO function and KEGG pathway enrichment analysis on intersection targets through Metascape platform,and use Autodock vina software for molecular docking Simulation verification and visualization of the results.Research results:(1)According to the inclusion and exclusion criteria,a total of 145 prescriptions were included in the CAG medical records,including 162 drugs,30 drugs with a frequency ≥ 20,and the top 10 drugs were Jiao Sanxian,Pinellia,Taizishen,Magnolia,Corydalis,Inula,Coptidis,Hedyotis diffusa,octopus,Curcuma;among the four qi,warm,flat,and cold drugs are used more;among the five flavors,the frequency of bitter drugs is the highest,followed by sweet and pungent;The spleen,stomach,lung,and liver are the most common meridians of the drug;the efficacy analysis mainly focuses on tonic,phlegm,heat-clearing,blood-activating,and qi-regulating drugs;the ones with higher correlation strength are "Coptis chinensis,Taizishen,Jiao Sanxian-Pinellia","Corydalis,TaizishenPinellia",etc;complex network analysis shows that the core formulations are Jiao Sanxian,Pinellia,Taizishen,Magnolia officinalis,Corydalis,Inula,Coptis chinensis,Hedyotis diffusa.(2)Through the analysis of core prescriptions by network pharmacology,114 active ingredients,1276 targets,862 disease targets were obtained,and 229 intersection targets were obtained,among which the main compounds were quercetin,berberine,luteolin Herbalin,vitamin E,etc;the key targets in the PPI network are TNF,IL6,ALB,TP53,etc;the intersection genes are enriched in 204 molecular functions,2475 biological processes,132 cellular components,206 KEGG pathways,the key pathways are PI3K/AKT signaling pathway,NF-κB signaling pathway,cancer pathway,Th17 cell differentiation,apoptosis,etc.Molecular docking shows that the binding energies of active ingredients and targets are all≤-5.0k J/mol,with good binding active.Conclusions:(1)The data mining results show that Professor Mi Ziliang’s treatment of CAG is mainly based on the drugs that are sweet,pungent,warm,bitter and cold,and return to the spleen,stomach,lung and liver meridians.Most of them have the functions of tonic,phlegm,heat-clearing,blood-activating,and qi-regulating,which can reduce phlegm-heat,regulate qi and blood,and harmonize stomach qi,so that the spleen does not help the evil,and the evil does not hurt the positive;the core prescriptions are Corydalis,Inula,Taizishen,Jiao Sanxian,Coptis chinensis,Magnolia,Pinellia,Hedyotis diffusa.The whole recipe collectively embodies the treatment methods such as the combination of cold and heat,supplementation of replenishment and reduction,appropriate lifting and lowering,simultaneous application of elimination and replenishment,and harmony of qi and blood.The word "harmony" runs through it.(2)Through network pharmacology research,the mechanism of action of core prescriptions in the treatment of CAG may be the active ingredients quercetin,berberine,luteolin,vitamin E,etc,through affecting TNF,AKT1,IL6,ALB,TP53 and other targets,to regulate PI3K/AKT signaling pathway,NF-κB signaling pathway,cancer pathway,Th17 cell differentiation,cell apoptosis,etc.Molecular docking shows that luteolin and vitamin E may have therapeutic effects on diseases by acting on the receptor ALB.
