Objective: JARID1B(Jumonji,A T rich interactive domain 1B)is a histone 3 subunit 4 lysine(H3K4)specific demethylase.JARID1 B plays an important role in cell renewal,differentiation and some biological changes.JARID1 B is up-regulated in oral,thoracic,lung,bladder,prostate,hypopharyngeal and other malignant tumors.At present,JARID1 B is considered to be related to tumor growth and cancer cell proliferation,and may be involved in cell cycle metabolism or regulation.β-Catenin is also called β-catenin.As a regulatory protein,β-Catenin plays an important role in WNT signaling pathway.In resting cells,β-Catenin binds to intracellular E-cadherin(E-Cadherin)to stabilize intercellular adhesion.Under the activation of WNT signal,β-Catenin,as a transcriptional activator,enters the nucleus and participates in the activation of a variety of downstream signal pathways,and can directly or indirectly regulate the biological processes such as cell division,differentiation,metabolism and apoptosis.In recent studies of hypopharyngeal and colorectal cancer,it has been found that JARID1 B can promote the expression of β-Catenin.It is speculated that JARID1 B may play a regulatory role through WNT/ β-Catenin signal pathway.In this study,we detected the expression of JARID1 B and β-Catenin in nasopharyngeal carcinoma tissue / normal tissue,analyzed the difference of their positive expression rate and expression degree between cancerous tissue and normal tissue,and discussed the relationship between them and clinical stage,T stage,cervical lymph node metastasis and prognosis of nasopharyngeal carcinoma,so as to provide basis for clinical search for new therapeutic targets.Methods:(1)the experimental group: 64 paraffin specimens of nasopharyngeal carcinoma diagnosed by nasopharyngeal biopsy were collected from August 2016 to August 2020 in the Department of Otorhinolaryngology,head and neck Department of Otorhinolaryngology of Qingdao University.Benign control group: 20 tissue paraffin specimens diagnosed as chronic nasopharyngitis were collected in the same period.The expression degree of the two groups of samples was quantified by immunohistochemical streptavidin peroxidase link staining and immunohistochemical scoring method.The expression degree equal to 0 is regarded as negative expression,the rest is positive expression,the expression degree above the median is high expression group,and below the median is classified as low expression group.(2)Statistics of clinical data of nasopharyngeal carcinoma group and follow-up of its outcome.(3)The data were processed by SPSS23.0 software to analyze the difference of positive expression rate and expression degree of JARID1 B and β-Catenin between nasopharyngeal carcinoma group and benign control group.To investigate the correlation between the positive expression rate and degree of JARID1 B and β-Catenin and clinical stage,T stage,cervical lymph node metastasis and prognosis in patients with nasopharyngeal carcinoma.Results: 1.Expression of JARID1 B in nasopharyngeal carcinoma group and benign control group:(1)JARID1B was positive in 64 cases of nasopharyngeal carcinoma and 20 cases of benign control group,and there was no difference between the them.(2)The positive expression rate of JARID1 B in nasopharyngeal carcinoma group was higher than those in benign control group,and there was significant difference between them.(Z=-2.047,P=0.041<0.05)2.Expression of JARID1 B in nasopharyngeal carcinoma:(1)The expression of JARID1 B has no significant relationship in terms of gender and age.(2)The expression of JARID1 B has no significant relationship in terms of clinical stage,T stage and cervical lymph node metastasis.(P>0.05).3.The relationship between JARID1 B and prognosis:(1)The expression of JARID1 B of high expression group / low expression group has no significant relationship in terms of survival time(follow-up)and progressionfree survival time.(P>0.05).(2)There was a significant correlation between the expression of JARID1 B and the time of survival(follow-up),which was a negative correlation.(P<0.05).There was no statistical relationship between the expression of JARID1 B and progression-free survival time.(P>0.05).4.Expression of β-Catenin in nasopharyngeal carcinoma group and benign control group:(1)In 64 cases of nasopharyngeal carcinoma,59 cases were positive and 5 cases were negative.In 20 cases of control group,12 cases were positive and 8 cases were negative.The positive rate in nasopharyngeal carcinoma group was higher than that in control group,and the difference was statistically significant.(χ2=9.734,P<0.05).(2)The expression of β-Catenin in nasopharyngeal carcinoma group was higher than that those in control group,and the difference was statistically significant(Z=-5.336,P<0.01).5.Expression of β-Catenin in nasopharyngeal carcinoma:(1)the expression of β-Catenin was no significant difference in terms of gender and age.(P>0.05).(2)The expression of β-Catenin in T3-T4 stage was higher than those in T1-T2 stage,and the difference was statistically significant.(Z=-2.010,P=0.044<0.05).The expression of β-Catenin has no statistical relationship in terms of clinical stage and cervical lymph node metastasis(P>0.05).6.The relationship between β-Catenin and prognosis:(1)The 3-year survival rate of nasopharyngeal carcinoma patients with positive expression of β-Catenin was 83.9%,which was higher than that of patients with negative expression(40%).There was a statistical relationship between them(P=0.031<0.05)There was no statistical relationship between the positive expression rate of β-Catenin and progression-free survival rate.(P>0.05).The expression of β-Catenin of high expression group / low expression group has no significant relationship in terms of survival time(follow-up)and progression-free survival time(P>0.05).(2)The expression degree of β-Catenin has no statistical relationship in terms of survival time(follow-up)and progression-free survival time.(P>0.05).7.Correlation between the expression of JARID1 B and β-Catenin in nasopharyngeal carcinoma:(1)There was no statistical relationship between the expression of JARID1 B and β Catenin in nasopharyngeal carcinoma.(P>0.05)Conclusion: 1.JARID1 B is highly expressed in nasopharyngeal carcinoma.There was a negative correlation between the expression of JARID1 B and the prognosis of patients.JARID1 B can be used as a potential tumor indicator of nasopharyngeal carcinoma and can be used to predict the prognosis of patients.2.β-Catenin showed high expression and high positive expression rate in nasopharyngeal carcinoma.β-Catenin is related to T stage of nasopharyngeal carcinoma,and the expression in T3-T4 stage was higher than that in T1-T2 stage.β-Catenin can be used as a potential tumor indicator of nasopharyngeal carcinoma and is related to the invasiveness of nasopharyngeal carcinoma.
