Background and ObjectiveIschemic stroke has brought great burden to society and families.For many years,researchers have been trying to find post-stroke treatment methods to improve the prognosis of stroke patients.However,many studies failed to transform.The reason may be that more study used rodents,which are to great difference from human in genetic,physiological and anatomical.Thus,Stroke Therapy Academic Industry Roundtable recommended to usd nonhuman primates in preclinical studies,for the reason that their organization structure,immune,physiological and metabolic are similar to humans and allowed for multiple invasive operation.As a special pattern of repetitive transcranial magnetic stimulation,theta burst stimulation,which has been widely used in neural regulation including post-stroke rehabilitation,simulates the endogenous shock of Theta frequency occurring in the cortex and hippocampus during learning,memory and movement.However,its mechanism remains unclear.Therefore,in this study,we constructed middle cerebral artery occlusion(MCAO)model in machin,and then TBS was used to intervene in MCAO machins.During the study,the differences of behavior,electrophysiology,and resting state functional magnetic resonance were compared in each time period too explore the possible mechanism of TBS in stroke therapy.Methods1.MCAO machins were constructed by electrocoagulation in healthy machins.The indexes of behavioral,imaging,electrophysiology and resting state functional magnetic resonance were collected and compared before and after operator.2.TBS was used in MCAO machins for 3 months and the informations of behavioral,electrophysiology and resting state functional magnetic resonance were collected and compared in TBS used for 0 month,1 months and 3 months.Some indexes were compared with pre-operation.Results1.All surviving MCAO machins showed stroke syndromes in behavioral,imaging and electrophysiology.The change of ALFF,f ALFF,Re Ho and functiona connectivity could be seen in resting state functional magnetic resonance before and after operation.2.Difference improvement could be seen in behavioral and electrophysiology after TBS.The change of ALFF,f ALFF,Re Ho and functiona connectivity could be seen in resting state functional magnetic resonance in difference periods.Some changes in rest-f MRI in some brain area such as precuneus after TBS was opposite to before-after operation.Conlusions1.When one hemisphere suffered from stroke,the unaffected hemisphers was affected and the activity of some brain areas,the intensity and amount of functional connectivity changed.2.TBS could influence the excitability of cerebral cortex,change the functional connectivity and finally regulates the whole brain function.
